摘  要:目的探讨IL-24基因联合大蒜素对肺癌A549细胞的增殖抑制和促凋亡作用。方法提取前期构建的p DC316-h IL-24-EGFP质粒,转染肺癌A549细胞,分为A组:空白对照组;B组:单独大蒜素组;C组:脂质体+IL-24组;D组:大蒜素+脂质体+IL-24。荧光显微镜下观察质粒的转染情况,结晶紫染色法和MTT法检测p DC316-h IL-24-EGFP联合大蒜素对肺癌A549细胞生长的影响,用流式细胞仪检测细胞凋亡。结果 40、80 mg/L大蒜素和p DC316-h IL-24-EGFP单用24、48 h对A549细胞均有明显抑制作用,呈剂量依赖性。低剂量大蒜素(40 mg/L)联合p DC316-h IL-24-EGFP比单用p DC316-h IL-24-EGFP作用更强(P<0.05)。40 mg/L大蒜素、p DC316-h IL-24-EGFP和两药联用48 h,A549细胞凋亡率分别为32.7%,39.4%和68.8%。结论 p DC316-h IL-24-EGFP联合大蒜素对人肺癌A549细胞有协同抑制作用。Objective To investigate the inhibitory effect of combined treatment of IL-24 gene with allicin on A549 lung cancer cell lines. Methods Pre-constructed recombinant plasmid pDC316-hIL-24-EGFP was extracted and tranfected to A549 cells. There were four groups:Group A( blank control) ,Group B( allicin) ,Group C( pDC316-hIL-24-EGFP with lipofectamine transfection),Group D(pDC316-hIL-24-EGFP with lipofectamine transfection combined with allicin) . The transduction efficiency of target cells was detected by fluorescent microscope. Cytotoxic effects of pDC316-hIL-24-EGFP combined with allicin on A549 was detected by crystal violet staining and MTT assay. Results Either allcin (40 mg/L or 80 mg/L) or pDC316-hIL-24-EGFP could inhibit the growth of A549 lung cancer cells in a dose dependent manner. Combined treatment of allcin 40 mg/L with pDC316-hIL-24-EGFP resulted in asynergistic effect of inhibiting cell growth(P〈0. 05). After incubation with allcin 40 mg/L,pDC316-hIL-24-EGFP and in combination,the apoptosis rate of A549 cells was 32. 7%,39. 4% and 68. 8%,respectively. Conclusion The combination of pDC316-hIL-24-EGFP and allcin has a synergistic effect on the growth of A549 cells.
