摘 要:目的:研究不同浓度的AAVC-Ⅰ对人口腔鳞癌Tca8113细胞GRP78、Caspase-4基因表达的影响。方法:设置正常对照组和AAVC-Ⅰ实验组,Trizol试剂提取各组总RNA,运用RT-PCR法扩增内参基因β-actin及目的基因GRP78、Caspase-4片段,Chemi Doc XRS凝胶成像系统观察电泳结果并采用Quantity One软件测定目的条带灰度值。结果:正常对照组、实验组均扩增出GRP78、Caspase-4目的条带,且随AAVC-Ⅰ浓度增大GRP78、Caspase-4表达增加。结论:高浓度AAVC-Ⅰ可上调GRP78、Caspase-4基因的表达,因此AAVC-Ⅰ可能诱导人口腔鳞癌Tca8113细胞发生了内质网应激。Objective: To observe the effects anti-tumor component-Ⅰ from Agkistrodon acutus venom( AAVC-Ⅰ) in diverse dosage on the expression GRP78 and Caspase-4 gene in human oral squamous carcinoma Tca8113 cell lines. Methods: The experiment was performed in normal control group and AAVC-Ⅰ group. Trizol reagent was used to extract total RNA in each group,and RT-PCR was performed to amplify the internal control β-actin gene and fragments of GRP78 and Caspase-4. Chemi Doc XRS gel imaging system was used to observe electrophoresis presentation,and Quantity One software to measure the gray value in the target band. Results: Both GRP78 and Caspase-4 were amplified,and the expression of these two genes was up-regulated with increased dose of AAVC-Ⅰ. Conclusion: High dose of AAVC-Ⅰ can increase the expression of GRP78 and Caspase-4 genes,suggesting that AAVC-Ⅰmay induce endoplasmic reticulum stress in the oral squamous carcinoma Tca8113 cell lines.
