摘 要:目的对比细胞悬液法及组织块包埋法制作兔间变表皮鳞癌VX2瘤株肌肉肿瘤模型差异,选择较为合理的造模方法。方法取传代VX2肿瘤荷瘤兔,将肿瘤制备成肿瘤组织块及细胞悬液,将瘤块包埋于10只新西兰兔左腿股外侧肌内(组织块包埋组)、将肿瘤细胞悬液注入此10只家兔右腿股外侧肌内(细胞悬液组)制备VX2肌肉肿瘤模型。于模型制备后第7、14、21天行彩色超声检查,观察肿瘤形态、测量肿瘤体积;模型制备后第21天处死动物,取肿瘤测量实际体积。结果2种方法建模成功率均为100%。建模后第7、14、21天超声测量组织块包埋组肿瘤体积均大于细胞悬液组[(0.13±0.07)cm3比(0.05±0.01)cm3,(1.66±0.59)cm3比(0.94±0.30)cm3,(13.30±4.68)cm3比(7.15±2.65)cm3],差异均有统计学意义(均P〈0.05)。模型制备后21d,大体观察组织块包埋组肿瘤较规则,无异位种植,细胞悬液组肿瘤形状不规则,可见肿瘤异位种植;模型制备第21天超声测量肿瘤体积与实际测量肿瘤体积差异无统计学意义[组织块包埋组:(13.30±4.68)cm3比(12.20±4.86)cm3,细胞悬液组:(7.15±2.65)cm3比(6.42±2.33)cm3](P〉0.05)。结论瘤块包埋法较细胞悬液法更为方便,不易发生异位种植,是一种理想的肌肉肿瘤模型制备方法。Objective To compare the cell suspension method and tumor tissue embedding method in establishment of VX2 muscle tumor model in rabbits. Methods After separating the VX2 tumor from cancer bearing VX2 rabbit, the tumor tissue blocks and cell suspension were prepared; the tissue blocks was embedded into the left vastus lateralis (tissue block group) and the cell suspension was injected into the right vastus lateralis (cell suspension group) of 10 rabbits. Color Doppler ultrasound was used to observe the tumor and measure the tumor volume 7, 14 and 21 d after model establishment. The rabbits were sacrificed and the tumors were taken out for volume measurement 21 d after model establishment. Results The success rates were both 100% for two methods. The tumor volume in tissue block group was significantly greater than that in cell suspension group 7, 14 and 21 d after model establishment [ (0. 13 ±0. 07) cm3 vs (0. 05 ±0. 01 ) cm3, (1.66 ±0. 59) cm3 vs (0. 94 ± 0. 30) cm3, (13.30 ±4. 68) cm3 vs (7. 15 ±2. 65) cm3] (P 〈0. 05). Twenty one days after model establishment, in gross observation, the tumor sharp was regular and no ectopic implantation was observed in tissue block group, while the tumor sharp was irregular and ectopie implantation was observed in cell suspension group; compared with actual tumor volume, the volume measured by ultrasound show no significant difference [ tissue block group: (13.30±4.68) cm3 Vs (12.20±4.86) cm3, cell suspension group: (7.15 ±2.65) cm3 vs (6.42 ± 2. 33 ) cm3 ] ( P 〉 0.05 ). Conclusion The tissue embedding method is more convenient with less eetopic implantation in the establishment of muscle tumor model compared with cell suspension method.
