摘 要:目的研究解聚素-金属蛋白酶17(ADAM17)在胃癌中的表达及其对胃癌细胞增殖及侵袭能力的影响。方法收集60例手术切除的胃癌组织和20例正常胃黏膜组织,采用免疫组化sP法检NI]ADAM17在胃癌及正常胃黏膜组织中的表达情况,分析ADAM17在胃癌中的表达与胃癌患者临床病理特征的关系。利用脂质体介导的ADAM17-shRNA转染人胃癌细胞SGC-7901,采用实时荧光聚合酶链反应(RT-PCR)和Westernblot检测转染沉默效率;以CCK-8细胞增殖实验、Transwell小室试验观察通过抑制ADAM17基因表达对人胃癌细胞增殖及侵袭的影响。结果ADAM17蛋白在胃癌组织中表达阳性率为78.33%,在正常胃黏膜组织中表达阳性率为15.00%,差异具有统计学意义(P〈0.01)。胃癌患者ADAM17蛋白阳性表达与肿瘤浸润深度、TNM肿瘤分期有明显相关性,而与患者的年龄、性别、肿瘤大小、淋巴结转移、远处转移情况无关。SGC-7901细胞成功转染特异性ADAM17-shRNA后,通过RT-PCR和Western blot检测显示,ADAM17基因的表达水平明显抑制;同时SGC-7901细胞增殖侵袭能力也明显下降,与未转染组和阴性对照组比较差异有显著性(P〈0.05)。结论ADAM17在胃癌组织中呈高表达,ADAM17-shRNA通过脂质体转染人胃癌细胞SGC.7901抑制ADAM17蛋白产物表达后,细胞增殖侵袭能力明显下降,提示ADAM17基因在胃癌进展过程中可能起到重要作用。Objective To evaluate the expression of a disintegrin and metalloproteinase 17 (ADAM17) protein in patients with gastric cancer, and to observe the effects of ADAM17 on proliferation and metastasis of human gastric carcinoma cell. Methods SP immunohistochemisty was applied to detect the expression of ADAM17 in 60 cases of gastric cancer, 20 normal gastric mucous membrane. ADAM17 shRNA was transfected by lipidosome into SGC-7901, and then we used RT-PCR to check ADAM17 mRNA expression; ADAM17 protein was detected by Western blot essay; CCK-8 and Transwell chamber assay were performed to detect the effects of ADAM17 on cell proliferation and metastasis of SGC-7901. Results The expression of ADAM17 in gastric cancer was higher than in normal gastric mucous membrane (P 〈 0.01). Significant relationship was found between the expression of ADAM17 and the depth of tumor invasion and staging of carcinoma. The expression of ADAM17 mRNA and protein in SGC-7901 was decreased with ADAM17-shRNA transfected. A dramatic decline was found in proliferation and metastasis of SGC-7901 which was transfected. Conclusion ADAM17 is highly expressed in gastric cancer tissue, and its overexpression status is closely related with the ability of tumor proliferation and metastasis.
