摘  要:目的研究lncRNA H19遗传变异与肝癌易感性的关联,利用生物信息学的方法在实验前期筛选出H19中有潜在功能的SNPs。方法运用1000 Genomes Project和Ensembl等数据库确定H19 SNPs名录,利用Allele frequency calculator选取MAF值大于0.05的SNPs;结合文献查阅,运用SNP Function Prediction和F-SNP等功能预测软件筛选有潜在功能的SNPs。结果 5个SNPs在转录调控、蛋白质编码以及剪切调控中可能存在相应生物活性,值得进一步研究。结论对lncRNA H19的SNPs进行了科学合理的选择,可以更全面深入地探寻H19影响肝癌发生发展的遗传机制。Objective To study the association between lncRNA H19 genetic variation and liver cancer susceptibility,we screen out the potential SNPs of H19 by comprehensive bioinformatics strategy. Methods1000 Genomes Project,Ensembl and other databases were applied to query the list of SNPs. Allele frequency calculator was used to select the SNPs with the MAF values 0.05. SNP Function Prediction and F-SNP were utilized to predict the function of SNPs. Results 5 SNPs had been identified with potentially biological activity in transcriptional regulation,protein coding and regulation of splicing,which should be further studied. Conclusion SNPs in IncRNA H19 may be scientifically and reasonably collected,which provides an evidence for exploring the effect of H19 on the occurrence and development of liver cancer.
