摘　　要：目的检测β-ccatenin在原发中枢神经系统淋巴瘤（PCNSL）患者组织中的表达。方法收集2010年10月至2012年4月诊断为PCNSL的10例患者的蜡块组织（实验组），同时收集10例淋巴结炎患者的蜡块组织作为对照组。分别采用实时定量聚合酶链反应（PCR）和免疫组织化学方法检测β-ccateninmRNA和蛋白的表达，分析其表达与临床病理因素的关系。结果免疫组织化学显示B．catenin定位于细胞质和（或）细胞膜，10例PCNSL组织中阳性表达4例，相对于内参基因，目的基因相对表达量为4．70±0．57，对照组织中无阳性表达者，相对表达量为1．00±0．27，两组阳性率差异有统计学意义（P〈0．05）。β-ccatenin的表达与患者年龄、功能状态（PS）评分、脑脊液蛋白水平和血清乳酸脱氢酶（LDH）水平无关（均P〉0．05）。结论无论在mRNA水平还是蛋白质水平，β-ccatenin在PCNSL组织中均高表达，且有细胞质内的表达，而淋巴结炎组织中无此种现象。[Abstract] Objective To detect the expression of β-ccatenin in the tissues of primary central nervous system lymphoma （PCNSL）, and to discuss its function in PCNSL. Methods The paraffin embedded tissues from 10 patients diagnosed as PCNSL from October 2010 to April 2012 were collected as the experimental group. The paraffin embedded tissues from 10 patients with lymphadenitis were collected as the control group. Quantitative real-time PCR and immunohistoehemical method were used to detect the expression of β-ccatenin in these tissues, and the relationships between β-ceatenin and clinical data were analyzed. Results Immunohistochemistry results showed that β-ccatenin protein was localized in the cytoplasm and （or） nucleus. Among 10 PCNSL patients, β-ccatenin protein was positive in 4 patients, while it was no positive in all of 10 lymphadenitis patients, with the significant differences between both groups （P 〈 0.05）. The β-ceatenin gene relative expression level was 4.70±0.57 and 1.00±0.27 in the experimental group and the control group, respectively. β-ccatenin expression was no correlation to age, PS score, cerebrospinal fluid protein level and serum lactate dehydrogenase level of patients with PCNSL. Conclusions Whether in mRNA level or in protein level,β-ccatenin expression is always high in PCNSL tissues, and its protein is expressed in the cytoplasm, however, this phenomenon was not observed in the tissue of lymphadenitis.
