头孢雷特对细菌感染小鼠败血症的体内抗菌作用

摘　　要：目的观察头孢雷特对细菌感染败血症小鼠的体内抗菌作用,并与头孢呋辛钠、头孢孟多酯钠进行比较。方法腹腔注射最低致死量的细菌混悬液0.5mL,建立小鼠败血症模型。将三种药物各设5个剂量组(6.3,12.5,25.0,50.0,100.0 mg·kg^(-1)),每组10只小鼠。于造模后1 h,静脉注射不同浓度的抗菌药物0.2 mL。观察感染鼠死亡率(共14 d),计算感染保护作用的50%有效剂量(ED_(50)值)。结果头孢雷特、头孢呋辛钠和头孢孟多酯钠对甲氧西林敏感金黄色葡萄球菌感染小鼠体内保护作用的ED_(50)值分别为32.59,40.27,34.65 mg·kg^(-1),对超广谱β内酰胺酶阴性(ESBL^-)大肠埃希菌感染小鼠体内保护作用的ED_(50)值分别为32.57,39.78,34.70 mg·kg^(-1),对ESBL-肺炎克雷伯菌感染小鼠体内保护作用的ED_(50)值分别为35.36,37.72,34.88 mg·kg^(-1),对变形杆菌感染小鼠体内保护作用的ED_(50)值分别为26.80,37.12,32.79 mg·kg^(-1)。三种药物的ED_(50)值比较差异无统计学意义(P>0.05)。结论头孢雷特体内抗菌作用与头孢呋辛钠、头孢孟多酯钠相当。Objective To evaluate the in vivo activity of ceforanide in the treatment of mouse septicemia and compare its effect with that of cefuroxime sodium and cefamandole sodium. Methods The mouse septicemia model was established by an intraperitoneal injection of the suspensions（ minimal lethal dose） of bacteria 0. 5 mL. Each antibiotic was tested in five concentration groups（ 6. 3, 12. 5, 25. 0, 50. 0, 100. 0mg·kg^（-1）） and each group consisted of 10 mice. Ceforanide,cefuroxime and cefamandole 0. 2 mL were injected to the mice in 1 h after bacteria injected. The death rate in each group was monitored in 14 days. The 50%of animals protected dose（ ED_（50）） was calculated. Results ED_（50） values of ceforanide,cefuroxime sodium and cefamandole sodium against Methicillin- sensitive Staphylococcus aureus were 32. 59,40. 27,34. 65 mg·kg^（-1）,against extended- spectrum beta- lactamase negative（ ESBL^-）Escherichia coli infection were 32. 57,39. 78,34. 70 mg · kg^（-1）,against ESBL-Klebsiella pneumoniae infection were 35. 36,37. 72,and 34. 88mg·kg^（-1）,against Proteus were 26. 80,37. 12,32. 79 mg·kg^（-1）. There was no statistically significant difference between the three antibiotics in ED_（50）（ P 〉0. 05）. Conclusion Ceforanide,cefuroxime sodium,cefamandole sodium have same antibacterial effect in vivo.
