社交应激诱导C57BL/6小鼠抑郁样模型的建立及行为学评价

摘　　要：目的采用社交应激方式诱导C57BL/6小鼠建立抑郁样模型,并采用多种行为学方法对模型进行评价,以验证模型可靠性。方法将20只雄性C57BL/6(C57)小鼠采用随机数字表法分为正常对照组(正常组,10只)和应激模型组(模型组,10只)。另取CD1(ICR)小鼠(10只)用于社交应激。正常组未予干预措施,模型组每天给予社交应激,连续10天。采用社交接触实验、高架十字实验和悬尾实验对各组小鼠进行行为学评价;采用E LISA法检测小鼠血清皮质醇水平,评估模型可靠性。结果社交接触实验显示,与本组CD1小鼠不存在时比较,正常组小鼠社交区停留时间延长,角落区停留时间缩短(P<0.05),模型组社交区停留时间缩短,角落区停留时间延长(P<0.05);与正常组CD1小鼠存在时比较,模型组社交区停留时间减少,角落区停留时间增多(P<0.05)。与正常组比较,模型组进入开放臂、闭合臂及高架十字总进入次数明显减少(P<0.05),开放臂进入次数比例降低(P<0.05);模型组小鼠在悬尾实验中后4 min内不动时间延长(P<0.05);模型组血清皮质醇水平明显升高(P<0.01)。结论社交应激诱导C57小鼠建立社交应激性抑郁样模型较为可靠,可能适用于中西医结合病证模型的研究。Objective To establish social stress induced depression-like model in mice of C57BL/6 strain,and to assess its reliability using different behavioral methods.Methods Totally 20 male mice of C57BL/6 strain were divided into the normal group and the stress model group by random digit table,10 in each group.Another 10 CD1 mice were subjected to social stress.Mice in the normal control group received no stress,while those in the model group received social stress for 10 successive days.Behavioral assessment was performed using social interaction test（SIT）,the elevated plus-maze（EPM） test,tail suspension test（TST）,respectively.Serum Cortisol level was detected by ELISA to assess the reliability of the model.Results In the social interaction test when the social target（CD1 mice）was inexistent,mice in the normal control group spent longer time in the social interaction zone and less time in the corner zone（P 〈0.05）;mice in the model group spent less time in the social interaction zone and more time in the corner zone（P〈0.05）.Compared with the normal group when CD1 mice existed,mice in the model group spent less time in the social interaction zone and more time in the corner zone（P〈0.05）.Compared with the normal control group,the total times for entry into open arms,close arms,and the maze were obviously reduced（P 〈0.05）,and the proportion of entering open arms was significantly reduced（P 〈0.05） in the model group.In TST,the motionless time within the last 4 min was prolonged in the model group（P 〈0.05）.The serum Cortisol level in the model group was obviously elevated（P〈0.01）.Conclusion Social stress induced depression-like animal model in mice of C57BL/6strain was quite reliable and possibly suitable to be used in integrative medicine research of combination of disease and syndrome model.
