摘  要:目的阻断Wnt信号通路对多发性骨髓瘤骨病作用。方法收集30例诊断为多发性骨髓瘤患者和18例健康对照组的临床资料、血清;采用ELISA方法测定血清SDF-1,DKK-1水平;体外培养多发性骨髓瘤细胞株,分别加入35 ng/ml SDF-1和100 ng/ml DKK-1,通过Western Blot方法观察DKK-1的表达量;采用SPSS Statistics 13.0进行统计学分析。当P<0.05时认为存在统计学差异。结果 MM患者(n=30)血清SDF-1水平显著高于健康对照(n=18)[(3 724.5±1 586.8)pg/ml vs(3 028.6±587.4)pg/ml,P=0.015)]。MM患者血清DKK-1水平亦显著高于健康对照[(3 814.6±3 487.8)pg/ml vs(1 881.3±657.8)pg/ml,P=0.029)]。在体外培养MM细胞株,采用35 ng/ml SDF-1作用于MM细胞和100 ng/ml DKK-1作用于MM细胞,Western Blot结果 :显示在SDF-1介导下,DKK-1的表达量较对照组及SDF-1组都有明显升高(P<0.005)。结论 SDF-1与DKK-1共同介导多发性骨髓瘤。Objective Blocking the Wnt signaling pathway for multiple myeloma bone disease.Methods Collected clinical data and serum from 30 cases diagnosed as multiple myeloma and 18 cases as control; using ELISA method to exam the level of SDF-1,DKK-1 in serum; add 35 ng / ml SDF-1 and 100 ng /ml DKK-1 separately into the multiple myeloma cell lines cultured in vitro,then observe expression of DKK-1 through western blot; western blot results statistical analysis by SPSS Statistics 13. 0. There was difference in statistics when P〈 0. 05. Results SDF-1 level in serum of people diagnosed with MM( n = 30) was significantly higher than healthy control( n = 18)( 3724. 5 ± 1586. 8) pg / ml vs( 3028. 6 ± 587. 4) pg / ml,P = 0. 015). DKK-1in serum of MM was also has a significantly level than that of control( 3814. 6 ± 3487. 8) pg / ml VS( 1881. 3 ±657. 8) pg / ml,P = 0. 029). Added 35 ng / ml SDF-1 and 100 ng / ml DKK-1 separately into the multiple myeloma cell lines cultured in vitro,the results of western blot show that mediated by SDF-1,the expression of DKK-1 had a significantly improvement than the control and SDF-1 group( P〈 0. 005). Conclusions SDF-1 and DKK-1 mediated multiple myeloma together DKK-1.
