摘 要:目的:观察慢性食管内灌注盐酸是否可以诱导大鼠迷走神经相关的食管-支气管反射通路神经肽P物质表达发生改变,并进一步探索中医肺胃相关理论的物质基础。方法:将16只雄性SD大鼠,按随机数字表法分正常对照组(n=8)、盐酸组(n=8),盐酸组用戊巴比妥钠腹腔注射轻度麻醉大鼠,将6号胃管插入食管至食管中下端,灌注含0.5%胃蛋白酶的0.1 mol/L盐酸(8滴/min,20 min/d),连续灌注14 d。正常对照组用PBs代替盐酸灌注食管,提取食管、气管和肺组织标本进行病理学观察和P物质免疫组化法测定。结果:盐酸组与正常对照组相比,食道、胃及肺组织中P物质表达均明显增多(P<0.01)。结论:慢性食管内灌注盐酸可以诱导大鼠产生气道神经源性炎症,及食管-支气管反射通路发生P物质表达改变,提示P物质可能是中医肺胃相关理论的物质基础之一。Objective: To investigate if repeated intra- esophageal instillation of hydrochloric acid( HCl) may induce the change of neuropeptide in esophageal-bronchil reflex pathway associated with vagus nerve in rats and to explore the material basis of lung and stomach theory in traditional Chinese medcine. Methods: 24 male SD rats were randomized into2 groups: bland control group and HCl group.Animals in HCl group received instillation of 0.1 mol / L of HCl containing 0.5% pepsin at middle and lower parts of esophagus once a day for 14 days. The expressions of substance P( SP) in the airway,esophagus and Stomach were detected by immunohistochemistry. Results: The expressions of SP in the airway,esophagus and stomach in HCI group were significantly higher than those in bland control group( P〈0.05 and P〈0.01). Conclusion: Repeated intra-esophageal instillation of HCl can induce airway neurogenic inflammation and SP alternations in esophageal-bronchial reflex pathway in rats,which indicated that SP would be the material basis of lung and stomach theory in traditional Chinese medcine.
