黄芪多糖对沙眼衣原体重组MOMP免疫增效作用的研究

摘　　要：目的:探讨沙眼衣原体重组外膜蛋白(Chlamydia Trachomatis recombinant Major Outer Protein Ctr MOMP)对生殖道感染沙眼衣原体小鼠的免疫保护作用,并研究黄芪多糖(Astragalus Polysaccharide APS)对其的免疫增强效果。方法:在成功构建Ct-MOMP的原核表达系统的基础上提纯r MOMP。将BALB/C小鼠分为100μl PBS组、50μgr MOMP组、50μgr MOMP+50μg Al(OH)3组、50μgr MOMP+50μg APS组、50μg r MOMP+100μg APS组、50μg r MOMP+200μg APS组。其中PBS组为阴性对照组,加有Al(OH)3组为阳性对照组,共进行3次免疫,通过检测血清和生殖道局部抗体、培养生殖道脱落细胞以及制作生殖道病理切片来观察和分析小鼠感染情况。结果:成功构建Ct-MOMP原核表达系统并提纯出r MOMP。r MOMP有着明显的免疫保护作用,带佐剂的r MOMP免疫效果明显优于单一r MOMP,且中浓度的APS免疫增强效果明显优于低浓度和高浓度的APS,与Al(OH)3组没有明显差异。结论:r MOMP对生殖道Ct感染有着明显的免疫保护作用,且中浓度的黄芪多糖对r MOMP免疫增强效果最好。Objective： To determine the immunoprotective effect of recombinant expressed Ct MOMP protein（ r Ct-MOMP） in BALB / C mice whose genital tract was attacked by Ct. And to study immunologic enhancement of astragalus polysaccharide on r Ct-MOMP. Methods： We extracted the expressed r Ct-MOMP on the successfully established prokaryotic expression system of Ct-MOMP gene. BALB / C mice were divided into the groups： 100 μL PBS,50 μg r MOMP,50μg r MOMP＋50 μg Al（ OH）3,50 μg r MOMP＋50 μg APS,50 μg r MOMP＋100 μg APS and 50 μg r MOMP＋200 μg APS.The PBS group was regarded as negative control group and the group with Al（ OH）3regarded as positive control group.Mice were immuned three times in total. Through detecting the local antibodies of serum and genital tract,we cultivated the vaginal exfoliated cells and produced the genital tract pathology slice to observe and analyze the infections in mice. Results： The prokaryotic expression system of Ct- MOMP gene was established successfully and r Ct- MOMP was extracted.The r MOMP had obvious immune protection. RMOMP had obvious immune protection and the enhancing immune effection of r MOMP with adjuvant was better than that of the single r MOMP. And immune effection of the medium concentration APS was better than the high or low concentration,but had no significant difference compared with Al（ OH）3. Conclusions： The r MOMP produces obvious immune protection on mice whose genital tract was attacked by Ct and the medium concentration of astragalus polysaccharides enhancing r MOMP immune effect is the best.
