摘  要:目的:优选蜗牛酶降解茯苓多糖的反应条件。方法:以茯苓多糖酶解得率为指标,采用单因素试验针对影响茯苓酶解的pH、加酶率、固液比、反应温度、反应时间等5个主要因素分别加以研究,并利用正交试验进行优化。结果:优选茯苓多糖的降解条件为:pH=6,加酶率1.8%,固液比1∶20,酶解温度40℃,酶解时间5h。结论:在该优选条件下,茯苓多糖降解率为40.2%,证实蜗牛酶可有效降解茯苓多糖,提高茯苓多糖的水溶性,与传统的水提醇沉法比较,得率提高65%以上。Objective:In order to explore the optimal condition of Poria enzymatic hydrolysis,Methods:Snailase has been used through single-factor study and orthogonal experiment.The single-factors investigated includes:pH,enzyme rate,solid-liquid ratio,temperature and reaction time.With single-factor test results comprehensively considered,using L18(35)orthogonal arrangement test.Results:the optimum hydrolysis condition for Poria enzymatic hydrolysis:pH=6,1.8% of enzyme liquid ratio of 1:20,40℃as reaction temperature and 5hours as hydrolysis time,under which the yield of polysaccharides is 40.2%.Conclusion:it can effectively improve the water soluble of Poria cocos polysaccharides.Compared with the traditional Ethanol subsiding method,the yield increased by more than 65%.
