摘 要:从河北省某鸭场发病鸭中分离到一株Ⅲ型鸭肝炎病毒(DHV)GT株,采用组织切片、全基因组测序和动物回归试验对其进行鉴定。结果显示:该毒株接种1日龄雏鸭,能引起显著的临床症状和肝细胞病变,该毒株的LD50为105.2;全基因组测序显示:该毒株全长7 802 bp,含652 bp的5′非编码区和366 bp的3′非编码区,编码2 516个氨基酸的多聚蛋白。根据Blast比对发现,该毒株属于DHV-C,与中国分离毒株C-YCW(98.1%)的同源性最高;从进化关系来看,与韩国型毒株的亲缘关系最近。表明该毒株属于我国流行的新型鸭肝炎病毒,命名为DHV-HBGT株。A virus isolate GT obtained from a duckling with liver ascites in Hebei province was confirmed as duck hepatitis virus type Ⅲ (DHV-Ⅲ)by tissue seetion,whole-genome sequencing and animal experiments. The results showed as follows: the virus caused significant pathogenicity,hstopathology and liver cells lesions when it was inoculated into one-day-age ducklings with 0.2 mL, and LD50 was 10^5.2. The virus genome was 7 802 nucleotides long consisting of 5'- and T-terminal non-coding regions of 652 and 366 nucleotides respectively. The single open reading frame encoded a polypeptide of 2 516 amino acids. Comparative genome analysis with other available strains in GenBank indicated that GT strain shared 98.2% similarity at the nucleotide level with C-YCW that was also isolated in China. Phylogenetic analysis showed that it had the closest relationship with strains isolated in South Korea. GT strain would belong to the new type of duck hepatitis virus,named DHV-HBGT strain.
