摘 要:为工厂化生产的相关食品及农产品储存过程中微生物的鉴定及抑菌对策提供理论依据,采用高盐察氏液体培养基(Czapek Dox Broth)梯度盐浓度培养法,CDA(Czapek Dox Agar)及TSA(Trypticase Soy Agar)固体培养基进行分离纯化,16SrRNA基因测序,邻接法构建16SrRNA序列的系统发育树等方法,对工厂生产香肠肠衣腐烂黑斑中嗜盐细菌进行分离鉴定,研究其特征特性。结果表明:25%~30%盐浓度固体培养基中分离到3株嗜盐细菌(Gyfl1,Gyfl2,Gyfl3),经分子学分析鉴定Gyfl1为Halomonas variabilis,Gyfl2为Chromohalobacter israelensis,Gyfl3为Chromohalobacter canadensis;经药敏试验,庆大霉素、卡那霉素、利福平、红霉素和新霉素等常见抗生素均能抑制嗜盐菌株Gyfl1、Gyfl2和Gyfl3,而利福平抑菌效果更明显。The halophilic bacteria in decayed black spot of sausage casing were isolated and identified by the gradient salt concentration method(Czapek Dox Broth liquid medium),CDA(Czapek Dox Agar)and TSA(Trypticase Soy Agar)solid media,and the characteristics of the identified halophilic bacteria was analyzed by using 16 SrRNA gene sequencing and building phylogenetic tree of 16 SrRNAsequence to provide the theoretical basis for microbial identification and bacteriostat measures during the storage process of agricultural products and related foods produced by industrialization.Results:Three halophilic bacteria strains(Gyfl1,Gyfl2 and Gyfl3)are successfully isolated on the solid medium with 25% ~30%salt concentration. Gyfl1,Gyfl2 and Gyfl3 strains are identified as Halomonas variabilis,Chromohalobacter israelensis and Chromohalobacter Canadensis by molecular analysis respectively.The drug sensitive test shows that common antibiotics of gentamicin,kanamycin,rifampicin,erythromycin,and neomycin all can inhibit Gyfl1,Gyfl2 and Gyfl3strains but the antibacterial effect of rifampicin is the highest among common antibiotics.
