摘　　要：目的探讨Crk相关底物家族成员C20ORF32在结直肠癌(colorectal carcinoma,CRC)组织中的表达情况,分析其与临床病理学参数、细胞增殖指数和预后的关系。方法采用免疫组织化学(Elivision^(TM)plus法)检测组织芯片中,100例结直肠癌组织和80例癌旁正常结直肠黏膜组织中C20ORF32和Ki-67的表达情况;应用Western blot检测16例正常结直肠组织和结直肠癌组织中C20ORF32的蛋白表达水平。结果结直肠癌组织中C20ORF32的表达水平显著高于正常结直肠组织(P<0.001),C20ORF32蛋白的过表达与结直肠癌患者的TNM分(x^2=5.769,P=0.016)、淋巴结转移(x^2=5.023,P=0.025)和脉管侵犯(x^2=8.315,P=0.004)密切相关。此外,结直肠癌组织C20ORF32高表达与细胞的高增殖活性显著正相关(r=0.380,P<0.001)。C20ORF32蛋白过表达组总生存率(Overall Survival,Os)显著短于低表达组(x^2=9.548,P=0.002)。多因素Cox回归分析(Forward Stepwise法)显示,TNM分期(P=0.004)C20ORF32(P=0.025)是影响结直肠癌患者预后的独立危险因素。结论 C20ORF32的过表达在结直肠癌发生、发展过程中发挥了重要作用,可以作为预测患者预后的标志物。Objective To investigate the expression and significance of C20ORF32 in human colorectal carcinoma（CRC）.Methods The expression of C20ORF32 and Ki-67 in 100 cases of CRC tissues and 80 cases of adjacent normal tissues was detected by immunohistochemistry,Western-blot was used to detect C20ORF32 expression in 16 paired CRC and adjacent normal tissues.Results The expression level of C20ORF32 was significantly higher in human colorectal carcinoma than in normal colorectum（P〈0.001）.The overexpression of C20ORF32 was closely related to TNM stage（x^2 =5.769,P =0.016）,lymph node metastasis（x^2=5.023,P =0.025） and vascular invasion（x^2=8.315,P=0.004）.The overexpression of C20ORF32 was positively correlated to proliferation of CRC marked by Ki-67 staining in CRC（r=0.380,P〈0.001）.The prognosis of patients with overexpression of C20ORF32 protein was significantly worse than the patients with negative/ low expression（x^2 =9.548,P =0.002）.Multivariate COX regression analysis showed that both of TNM stage（p =0.004） and C20ORF32 overexpression were independent prognostic factors for patients with CRC.Conclusion C20ORF32 plays an important role in the development of CRC,and could be used as a biomarker for the prognosis of CRC.
