摘  要:目的探讨蝙蝠葛碱对成骨细胞分化成熟的影响及其机制。方法正常人成骨细胞株(NHOst)培养在含有100ng/ml的蝙蝠葛碱的培养基中,7d后,通过相差显微镜观察细胞形态的变化,通过碱性磷酸酶(alkaline phosphatase,ALPase)和硝酸银(Von Kossa)镀银染色检测钙沉积。通过RT-PCR分析核心结合因子A1(Core binding factor A1,Cbfa1)mRNA的表达。通过Western-blot检测骨基质蛋白Osteocalcin的表达。结果培养7d后,实验组细胞呈岛样生长并开始分化或熟,成熟的细胞形成骨结节,而对照组细胞则未见骨结节形成。通过碱性磷酸酶和硝酸银镀银染色发现,实验组细胞可见钙沉积,而对照组细胞未见钙沉积。实验组Cbfa1和Osteocalcin的表达都明显高于对照组。结论蝙蝠葛碱可促进骨细胞分化成熟。Objective To study the effect and possible mechanism of dauricine on the differentiation and maturity of osteoblast(NHOst).Methods The normal human NHOsts were cultured in medium with 100ng/ml of dauricine for seven days,the cell morphology was observed by phase contrast microscope,calcium deposition of the cells was detected by alkaline phosphatase(ALPase) and Von Kossa silver staining.The expression of core binding factor A1 mRNA(Cbfa1mRNA) was detected by RT-PCR.The expression of bone matrix protein osteocalcin was detected by Western-blot.Results After seven days cultivation,the cells grew in island shaped distribution and began to differentiate into mature cells in experimental group,mature cells formed bone nodules,but not formed bone nodules in control group.The calcium deposits were detected in experimental group,but not in control group by alkaline phosphatase and Von Kossa silver staining.The expression levels of core binding factor A1(Cbfa1) and osteocalcin(bone matrix protein) were higher in experimental group than in control group.Conclusion Dauricine can promote osteoblasts differentiation and maturity.
