摘  要:目的:为探讨表没食子儿茶素没食子酸酯(EGCG)体内和体外抗呼吸道合胞病毒(RSV)作用。方法:在体外以不同浓度的EGCG分别作用于RSV复制周期的3个阶段,用CPE法观察EGCG抑制RSV的致细胞病变作用,用MTT法检测EGCG作用RSV的病毒抑制率;在体内建立RSV感染的BALB/c小鼠的动物模型,通过观察肺外观和HE染色镜下观察、计算肺指数、实时定量RT-PCR检测RSV核酸水平和空斑试验检测肺组织匀浆病毒滴度,探讨EGCG对RSV感染的保护作用。结果:在体外,EGCG对RSV有抗生物合成作用和预防作用,但是无直接杀伤作用。在体内,口服EGCG能够减少RSV感染的BALB/c小鼠肺组织病变程度,肺指数、RSV核酸水平表达和病毒滴度较RSV组明显降低。结论:EGCG在体内外均具有较好的抗RSV感染与复制作用。Objective: To investigate the effects of epigallocatechin gallate (EGCG) against respiratory syncytial virus (RSV) in vitro and in vivo. Methods. The cell culture technique was used in Hep- 2 cells to get cell viability at different concentrations of EGCG by MTT assay. Cytopathic effect (CPE) and MTT were applied to observe the protective function of EGCG and it's ingredients were administered to cells in three different ways (administration before infection, administration upon infection, administration after infection) to treat the infectious model in vitro. The anti vi- ral activity in vivo was performed on BALB/c mice. The protective effect of EGCG on RSV infec- tion was investigated by HE staining, calculating lung index, detecting nucleic acid level of RSV and plaque assay detecting viral titers in lung tissue homogenates. Results. EGCG had antiviral biosynthesis on RSV infection and prevent viral adsorption to Hep-2 cells, other than direct viru-cidal effect in vitro tration of EGCG in replication in vitro The lung tissue damage and the viral titles of RSV decreased by oral adminis- the mice infected by RSV. Conclusion: EGCG may inhibit RSV infection and and in vivo.
