摘  要:目的建立耐甲氧西林金黄色葡萄球菌(MRSA)体外生物被膜(BF),研究不同p H值酸性氧化电位水(EOW)对其的杀灭作用。方法收集临床分离的MRSA,以硅胶膜为载体建立体外BF模型,用银染法观察BF形成情况,扫描电镜观察BF形态变化。p H〈3.0、p H 4.0~5.0和p H 6.0~7.0的EOW作用于BF,用连续稀释法对各组作用不同时间的存活菌进行计数,计算杀菌率,同时用荧光显微镜观察死菌量。结果 3组不同p H值的EOW作用5 min杀菌率均达99%以上,3组作用10 min和20 min后杀菌率均达99.99%,3组杀菌率比较差异无统计学意义(P〉0.05);荧光显微镜观察显示,不同p H值EOW作用5、10、20 min后死菌数量明显增加,但3组间死菌量差异不明显。扫描电镜观察显示,随着EOW作用时间延长,BF出现断裂、细菌形态变皱甚至破碎更明显。结论不同p H值的EOW对MRSA体外BF均具有较好的杀灭作用,p H 6.0~7.0的EOW无腐蚀性,推荐将其用于临床MRSA感染的创面、伤口、医疗器械消毒。Objective To investigate the killing effect of electrolyzed oxidizing water(EOW)of different p H values on the extracorporeal biofilm(BF)of methicillin-resistant staphylococcus aureus(MRSA).Methods Collecting MRSA isolated clinically,the extracorporeal BF mode of MRSA was established with silica gel membrane as the carrier.The BF forming condition was observed by silver staining method,and the mophological change of BF was observed by scanning electron microscope.Acting on the BF with EOW of p H 3.0,p H 4.0-5.0 and p H 6.0-7.0,the viable bacteria of different acting time and sterilization rate were counted by continuous dilution method,and the amount of dead bacteria was observed simultaneously by fluorescence.Results After acting on BF of MRSA with EOW of three different p H levels,the sterilization rates on MRSA BF were all more than 99% for 5min,99.99% for 10 min and 20 min.There were no significant differences in sterilization rates among three different p H levels(P 0.05).Fluorescence microscopy observation showed that after acting on BF of MRSA with EOW of three different p H levels,the amount of dead bacteria obviously increased at 5min,10 min and 20 min,while there were no significant differences among three different p H levels.Scanning electron microscope observation showed that the BF fracture appeared and the bacteria shapechanged as the appearance of crumpling even obvious break with the prolongation of acting time of EOW.Conclusions The acidic EOW of different p H values all has strong eradicating effect on extracorporeal BE of MRSA,however,the EOW of 6.0-7.0 p H levels is recommended for disinfecting the surface of wound,wound and medical apparatus because of no corrosivity.
