摘 要:目的研究帕瑞昔布对非小细胞肺癌细胞株A549细胞黏附能力和CD44v6表达的影响,探讨其对肿瘤细胞转移行为的可能影响及机制。方法体外培养A59细胞,取对数生长期的A549细胞,采用随机数字表法将其分为4组:对照组(C组),10μmol/L帕瑞昔布组(P_1组)、40μmol/L帕瑞昔布组(P_2组)和160μmol/L帕瑞昔布组(P_3组)。C组细胞正常培养,P_1、P_2和P_3组细胞分别加入10、40和160μmol/L浓度的帕瑞昔布处理A549细胞24 h。纤维连接蛋白包被法检测各组细胞的黏附率,Western blot法检测各组细胞CD44v6的表达情况,其水平以条带灰度值/内参灰度值表示。结果与C组A549细胞的黏附率[(90.52±6.62)%]比较,P_1组[(77.38±4.93)%]、P_2组[(68.22±5.71)%]和P_3组[(55.01±6.24)%]依次降低,呈剂量依赖关系(P<0.05);与C组A549细胞CD44v6蛋白的表达水平(0.81±0.15)比较,P_1组(0.54±0.13)、P_2组(0.39±0.09)和P_3组(0.15±0.04)水平依次下调,呈剂量依赖关系(P<0.05)。结论帕瑞昔布可以降低非小细胞肺癌细胞株A549的黏附率,其机制可能与下调CD44v6的表达有关。Objective To study the effect of parecoxib on adhesion ability of non-small cell lung cancer cell line A549 and CD44v6 expression in A549 cells and explore its potential influence on metastatic behavior of tumor cells and the mechanism.Methods After being cultured in vitro A549 cells in logarithmic growth phase were divided into 4 groups by random number table method:control group(C group),10 μmol/L parecoxib group(P_1 group),40 μmol/L parecoxib group(P_2 group) and 160 μmol/L parecoxib group(P_3 group).The cells in C group were normally cultured.The cells in P_1,P_2 and P_3 groups were cultured in the culture medium containing 10,40 and 160 μmol/L parecoxib,respectively.After being cultured for 24 hours,cell adhesion rate was detected by fibronectin coated method,and CD44v6 relative expression level(ratio of band gray-scale value to internal reference gray-scale value) was detected by Western blot method.Results Compared with C group[(90.52 ±6.62)%],the adhesion rate of A549 cells in P_1 group[(77.38 ±4.93)%],P_2group[(68.22 ± 5.71)%]and P_3 group[(55.01 ± 6.24)%]decreased in turn with dose-dependent manner(P 0.05).Compared with C group(0.81 ±0.15),the CD44v6 protein expression levels in P_1 group(0.54 ±0.13),P_2 group(0.39 ± 0.09) and P_3 group(0.15 ± 0.04) were down-regulated in turn with dose-dependent manner(P 0.05).Conclusion Parecoxib can decrease the adhesion rate of A549 cells,and the mechanism may be associated with down-regulating the expression of CD44v6.
