摘 要:目的观察威草胶囊方中健脾补肾药和祛湿活血药对尿酸性肾病大鼠肾组织结缔组织生长因子(CTGF)和肝细胞生长因子(HGF)表达的影响,探讨威草胶囊方中不同组分保护肾功能的具体机制。方法将60只清洁级Wistar雄性大鼠随机分为正常组、模型组、别嘌呤醇组、威草胶囊组、健脾补肾组和祛湿活血组,每组10只。用酵母和腺嘌呤合用复制尿酸性肾病模型,复制成功后均改用普通饲料喂养。正常组和模型组每日给予蒸馏水2 m L灌胃,别嘌呤醇组给予别嘌呤醇0.05 g/(kg·d)灌胃,威草胶囊组给予威草胶囊混悬液1.08g/(kg·d)灌胃,健脾补肾组给予健脾补肾药混悬液0.58 g/(kg·d)灌胃,祛湿活血组给予祛湿活血药混悬液0.50 g/(kg·d)灌胃,共4周。用苏木精和伊红进行肾组织染色并制成病理切片,观察每组切片中大鼠肾组织的病理形态学,采用RT-PCR技术检测肾组织中CTGF mRNA及HGF mRNA的表达情况。结果模型组CTGF mRNA表达水平明显高于正常组(P<0.05),HGF mRNA表达水平明显低于正常组(P<0.05)。各给药组CTGF mRNA表达水平均明显低于模型组(P均<0.05),HGF mRNA表达水平均明显高于模型组(P均<0.05),威草胶囊组与别嘌呤醇组比较差异无统计学意义(P>0.05)。健脾补肾组和祛湿活血组CTGF mRNA表达水平均明显高于威草胶囊组(P均<0.05);HGF mRNA的表达水平健脾补肾组明显高于威草胶囊组(P<0.05),祛湿活血组明显低于威草胶囊组(P<0.05)。祛湿活血组CTGF mRNA、HGF mRNA表达水平均明显低于健脾补肾组(P均<0.05)。结论威草胶囊及其不同组分均可减轻肾间质纤维化程度,起到肾保护作用,机制可能与下调CTGF在尿酸性肾病大鼠肾组织中的表达,上调HGF表达有关。健脾补肾药与祛湿活血药合用对下调大鼠肾组织CTGF表达具有协同作用;健脾补肾药与祛湿活血药均有上调大鼠肾组织HGF表达的作用,而健脾补肾药上调大鼠肾组织HGF表达�Objective It is to observe the effect of invigorating spleen and kidney medicine and dispelling dampness and promoting the circulation of blood medicine included in separated formula for Weicao capsule on the expressions of connective tissue growth factor( CTGF) and hepatocyte growth factor( HGF) in the renal tissues of uric acid nephropathy rats by separating formula,so as to investigate the specific mechanism of protecting renal function of different components of Weicao capsule recipe. Methods 60 specific pathogen free male wistar rats were randomly divided into six groups: normal control group,model group,allopurinol group,Weicao capsule group,invigorating spleen and kidney medicine group and dispelling dampness and promoting the circulation of blood medicine group. Uric acid nephropathy model was reproduced by using yeast and adenine.Then the rats was fed with normal diet. At last,every group was given corresponding processing. The normal control group was administered with distilled water( 2 m L / d) by gavage. The allopurinol group was administered with adenine at a dose of 0. 05 g /( kg·d) by gavage. The Weicao capsule group was given Weicao capsule suspension at a dose of 1. 08 g /( kg·d) by gavage. The invigorating spleen and kidney medicine group was given invigorating spleen and kidney medicine suspension at a dose of 0. 58 g /( kg·d) by gavage. The dispelling dampness and promoting the circulation of blood medicine group was given dispelling dampness and promoting the circulation of blood medicine suspension at a dose of 0. 50 g /( kg·d) by gavage. The treatment was lasted for four weeks. Expressions of CTGFmRNA and HGFmRNA were assessed by real time fluorescent quantification RT- PCR. Results The expression of CTGFmRNA of model group were significantly raised and the expression of HGFmRNA was significantly reduced compared with normal control group( P 〈0. 05).
