摘 要:目的观察丹参单体IH764-3对糖尿病肾病大鼠肾脏水通道蛋白(AQP)2、AQP4及肺脏AQP1表达的影响,探讨丹参单体IH764-3对糖尿病肾病大鼠水液代谢异常的调节机制。方法从60只SD大鼠中随机选择10只作为正常组,剩余50只采用高糖高脂饲料刺激加小剂量STZ诱导糖尿病肾病大鼠模型,将成模大鼠30只随机分为模型组、对照组、研究组各10只。对照组给予卡托普利灌胃,研究组给予卡托普利灌胃同时腹腔注射丹参单体IH764-3,正常组和模型组灌胃等量蒸馏水。灌胃16周末处死大鼠,留取肾、肺组织行病理观察,采用免疫组化法、Western blot印记法、RT-PCR法检测肾组织中AQP2、AQP4及肺组织中AQP1蛋白及mRNA表达情况。结果肾组织中AQP2、AQP4蛋白及mRNA表达量模型组均明显高于正常组(P均<0.05),对照组、研究组均明显低于模型组(P均<0.05),研究组均低于对照组(P均<0.05)。肺组织中AQP1蛋白及mRNA表达量模型组均明显低于正常组(P均<0.05),研究组与正常组比较差异无统计学意义(P>0.05),对照组与模型组比较差异无统计学意义(P>0.05)。结论 AQP2、AQP4在肾脏组织表达的增多及AQP1在肺脏组织表达的减少可能是导致糖尿病水液代谢异常的重要机制之一。丹参单体IH764-3对水通道蛋白的表达有调节作用,这可能是其调节水液代谢作用的机制,也是其防治糖尿病肾病的基础。Objective It is to observe the effect of expression of Salvia miltiorrhiza monomer IH764- 3 on the expression of Aquaporin 2( AQP2),AQP4 in kidney and AQP1 in lung in rats with diabetic nephropathy,so as to explore the the regulating mechanism of Salvia miltiorrhiza monomer IH764- 3 for body fluid metabolism abnormality. Methods 60 healthy SD rats were selected,among which ten as control group,and the other 50 ones were used to establish diabetes model by high glucose and lipoids feeder and low-dose STZ. They the successful models( n = 30) were randomly divided into 3 groups: model group,control group and study group,with 10 rats in each group. The control group was given captopril by gavage,study group was given captopril by gavage combined with IH764- 3 by intraperitoneal injection,normal group and control group were given distilled water at the same dose. After 16 weeks' treatment,the rats were killed to get their lung and renal tissue for pathological observation. The expression of protein and mRNA of AQP2,AQP4 in renal tissue and AQP1 in lung tissue were detected by immunohistochemistry method,Western blot method,RT- PCR method. Results The expression of protein and mRNA of AQP2 and AQP 4 in renal tissue in model group was higher than that in normal group( P〈0. 05),and that in control group and study group was lower compared with the model group( P〈0. 05),in study group was lower compared with control group( P〈0. 05). The expression of protein and mRNA of AQP1 in lung tissue in model group was lower than that in normal group( P〈0. 05),there was no significant difference in the indexes between study group and normal group( P〉0. 05),control group and model group( P〉0. 05). Conclusion The increase of AQP2 expression in renal tissue and decrease of AQP1 expression in pulmonary tissue may be one of the important mechanism which could induce body fluid metabolism abnormality.Salvia miltiorrhiza monomer IH764- 3 had regulatory effect on AQP expression,which may be the
