摘 要:目的 :探讨血小板型12-脂氧合酶(platelet type 12-lipoxygenase,p12-LOX)选择性抑制剂黄芩素(baicalein,BAI)对胃癌MKN-28细胞增殖和横向迁移的影响。方法:应用RT-PCR法检测胃癌MKN-28细胞和正常胃黏膜GES-1细胞中p12-LOX m RNA的表达。用不同浓度的BAI处理MKN-28细胞后,应用MTT法检测细胞的增殖情况,划痕愈合实验检测细胞的横向迁移能力,RT-PCR法检测细胞中p12-LOX m RNA的表达水平,免疫荧光染色法检测细胞中E-cadherin和Snail蛋白的表达。结果:胃癌MKN-28细胞中p12-LOX m RNA的表达水平高于正常胃黏膜GES-1细胞(P<0.01)。25、50和100μmol/L BAI处理1、3和5 d后,MKN-28细胞的增殖受到显著抑制,且具有浓度和时间依赖性(P值均<0.05)。25和50μmol/L BAI处理24和48 h后,MKN-28细胞的横向迁移能力受到抑制,且具有浓度依赖性(P<0.05)。25μmol/L BAI处理24和48 h后,MKN-28细胞中p12-LOX m RNA的表达水平均显著低于空白对照组(MKN-28细胞未进行任何处理)(P值均<0.01)。BAI处理组MKN-28细胞中E-cadherin蛋白的表达增强(P<0.01),而Snail蛋白的表达水平则降低(P<0.01)。结论:胃癌MKN-28细胞中p12-LOX m RNA呈过表达。BAI可显著抑制MKN-28细胞中p12-LOX m RNA的表达,并抑制细胞的增殖和横向迁移,其机制可能与E-cadherin和Snail蛋白的表达变化有关。Objective: To investigate the effects of platelet type 12-lipoxygenase(p12-LOX) inhibitor baicalein(BAI) on the proliferation and migration of gastric cancer MKN-28 cells.Methods: The expressions of p12-LOX m RNA in gastric cancerMKN-28 cells and normal gastric mucosa GES-1 cells were detected by reverse transcriptionPCR(RT-PCR). After MKN-28 cells were treated with different concentrations of BAI, the abilities of proliferation and horizontal migration were detected by MTT and wound-healing assay, respectively.The expression of p12-LOX m RNA was detected by RT-PCR, and the expressions of E-cadherin and Snail proteins were detected by immunofluorescence staining method.Results: The expression level of p12-LOX m RNA in MKN-28 cells was higher than that in normal gastric mucosa GES-1 cells(P 0.01). After treatment with 25, 50 and 100 μmol/L BAI for 1, 3 and 5 d, respectively, the proliferation of MKN-28 cells was significantly inhibited in a dose- and time-dependent manner(all P 0.05). After treatment with 25 and 50 μmol/L BAI for 24 and 48 h, respectively, the ability of horizontal migration of MKN-28 cells was inhibited in a dose-dependent manner(P 0.05). The expressions of p12-LOX m RNA in MKN-28 cells after treatment with 25 μmol/L BAI for 24 and 48 h were lower than those in the MKN-28 cells without any treatment(as a blank control)(both P 0.01). The expression of E-cadherin protein in MKN-28 cells in BAI treatment group was significantly increased(P 0.01), but the expression of Snail protein was decreased(P 0.01).Conclusion: p12-LOX m RNA is over-expressed in gastric cancer MKN-28 cells, and BAI can significantly inhibit the expression of p12-LOX m RNA and the proliferation and horizontal migration of MKN-28 cells. These effects may be related to regulation of expressions of E-cadherin and Snail proteins.
