摘 要:目的观察原花青素(proanthocyanidins,PAC)对血管性认知功能障碍(vascular cognitive impairment,VCI)大鼠认知功能和脑内单胺类神经递质和单胺氧化酶含量的影响。方法选取健康成年雄性Wistar大鼠48只,随机分为4组:假手术组(sham)、血管性认知功能障碍组(VCI)、低剂量原花青素组(L-PAC,100 mg/kg)和高剂量原花青素组(H-PAC,200 mg/kg)。双侧颈动脉结扎术制备血管性认知功能障碍模型,造模后每天灌胃给药,给药容量为2 ml,共给予28 d。Morris水迷宫检测VCI大鼠认知功能,水迷宫结束后取材,采用分光光度计检测前脑皮层单胺氧化酶(monoamine oxidase,MAO)的含量,高效液相色谱仪法检测大鼠前脑皮层去甲肾上腺素(norepinephrine,NE)、5-羟色胺(5-hydroxytryptamine,5-HT)和多巴胺(doparmine,DA)的含量。结果与sham组相比较,VCI组大鼠前脑皮层中5-HT、NE和DA的含量均显著下降(P<0.05),MAO含量明显升高(P<0.05);与VCI组相比较,低剂量和高剂量PAC治疗明显增加VCI大鼠前脑皮层中NE、DA和5-HT的含量(P<0.05),减少MAO的含量(P<0.05)。结论原花青素能够明显提高VCI大鼠的空间学习记忆能力,能够调节VCI大鼠脑内单胺类神经递质系统。Objective To explore the effects of proanthocyanidins( PAC) on the contents of monoamine neurotransmitters and monoamine oxidase( MAO) in vascular cognitive impairment rats. Methods Forty-eight healthy adult male Wistar rats were randomly assigned into four groups: sham group,VCI group,low-dose PAC group and high-dose PAC group. Bilateral common carotid arteries were occluded in healthy adult male Wistar rats to establish VCI model. Morris water maze was used to evaluate spacial learning and memory function. MAO was measured spectrophotometrically,and the levels of norepinephrine( NE),doparmine( DA) and 5-HT in the forebrain cortex were detected by HPLC. Results Compared to sham group,the contents of NE,DA and 5-HT significantly decreased in VCI group( P〈0.05),while MAO significantly increased( P〈0.05). Compared with VCI group,NE,DA and 5-HT significantly increased in both low-dose PAC group and high-dose PAC group( P〈0.05),while MAO decreased( P〈0.05). Conclusion PAC could significantly improve spacial learning and memory abilities,and restore central monoamine neurotransmitters system.
