摘 要:目的研究姜黄素和索拉非尼联合应用对人肾癌786-O细胞株增殖的影响。方法采用MTT法观察姜黄素和索拉非尼对人肾癌细胞786-O细胞株的抑制作用。结果姜黄素及索拉非尼能抑制人肾癌786-O细胞株的增殖,并呈浓度及时间依赖性,两者作用人肾癌786-O细胞株48 h的半数抑制浓度(IC50)分别为29.2μmol/L、6.5μmol/L。姜黄素6.25μmol/L、12.5μmol/L、25μmol/L、50μmol/L与索拉非尼1.5μmol/L、3μmol/L、6μmol/L、12μmol/L、联合24 h、48 h、72 h的抑制率显著高于单用索拉非尼、姜黄素组(P<0.05)。结论姜黄素在体外对人肾癌786-O细胞株的增殖有明显的抑制作用,能提高索拉非尼对人肾癌786-O细胞株的敏感性。Objective To explore the effects of curcumin combined with sorafenib on the proliferation of human renal carcinoma cell line-786-O in vitro. Methods The inhibitory effects of curcumin and sorafenib on growth of renal carcinoma cell line - 786-O were detected by MTT. Results The curcumin and sorafenib inhibited the proliferation of human renal carcinoma cell line -786-0 in a concentration and time-dependent manner, their IC50 was 29.2μmol/L,6.5μmol/ml on 48 hours, respectively. The combined inhibitory effects of cureumin at concentration of 6.25 μmol/L, 12.5 p, mol/L, 25μmol/L, 50 μmol/L and sorafenib at concentration of 1.5 μmol/L, 3 μmol/L, 6μmol/L, 12 μmol/L for 24h, 48 h, 72h were significantly higher than those of single application of curcumin or single application of sorafenib ( P 〈 0.05 ). Conclusion Curcumin can significantly inbibit the growth of 786-O cells, which can increase the sensitivity of 786-O cells to sorafenib.
