摘  要:目的观察大鼠肝癌细胞系N1经肝、脾原位接种后肝细胞癌(hepatocellular carcinoma,HCC)的发生与转移等情况,比较及筛选适宜的HCC N1发生及肺转移动物模型。方法 N1细胞进行体外培养。24只8周龄雌性BALB/c A裸鼠随机分为2组(12只/组)并分别进行N1的肝、脾原位接种,观察并比较两组间小鼠的存活率,肝、脾成瘤及肝、肺转移率等肿瘤生物及病理学特征。结果 N1细胞肝原位接种组肝成瘤率为100%,肿瘤呈结节状生长,与腹壁及周围组织有不同程度的粘连侵袭,肺及肺门淋巴结转移率分别为44%和22%,未见脾转移。N1细胞脾原位接种组脾成瘤率为64%,肝转移率为91%,肿瘤呈结节状生长,与腹壁及周围组织亦有不同程度的侵袭粘连,肺及肺门淋巴结转移率均为18%。结论 N1细胞肝、脾原位接种均易成瘤,是良好的HCC发生、发展及相关干预的动物模型,其中,肝原位接种鼠可能更适合于HCC发生与肺转移机制及药物干预等研究。Purpose To establish and compare liver cancer models in nude mice by injection of rat hepatocellular carcinoma (HCC) cells N1 via two different routes. Methods The HCC N1 cells were cultured in vitro first, and then injected into liver or spleen of fe- male BALB/eA nude mice. Thereafter, the mice survival rate, tumor formation rate, carcinogenic features, and metastasis were detec- ted. Results Mice that directly injected with N1 cells in liver were all developed liver cancer ( 100% ) , and presented an expansive growing pattern with surrounding invasion, 44% lung metastasis rate and 22% microscopically hilar lymph nodes metastasis. In the spleen orthotopic model, 64% animals developed HCC in spleen, with 91% liver metastatic rate, 18% lung and microscopically hilar lymph node metastasis rate. Conclusion This study providestwo valuable cancer models for the HCC carcinogenesis, and the direct liver injection mice is probably more useful for the studies of the underlying mechanism and therapeutic regimens for HCC metastasis.
