摘  要:目的探讨小分子干扰RNA(si RNA)下调Ezrin蛋白表达对人胃癌细胞SGC-7901侵袭迁移能力的影响及可能机制。方法采用脂质体法将Ezrin si RNA及阴性对照质粒分别转染SGC-7901细胞,q RT-PCR及Western blotting分别检测SGC-7901细胞中Ezrin m RNA及蛋白的表达情况,transwell实验检测细胞侵袭和迁移能力的变化,Western blotting检测YAP蛋白表达。结果转染Ezrin si RNA后,SGC-7901细胞Ezrin m RNA及蛋白表达水平显著降低(P<0.05),抑制了细胞侵袭和转移(P<0.05),YAP蛋白表达下调(P<0.05)。结论 Ezrin si RNA可抑制SGC-7901细胞迁移和侵袭,其机制可能与抑制YAP蛋白表达相关。Objective To explore the effect and mechanism of Ezrin down-regulation on the invasion and migration of gastric cancer cell line SGC-7901. Methods SGC-7901 ceils were transfected with Ezrin siRNA or negative control, qRT- PCR and Western blotting were performed to measure the expression of Ezrin mRNA and protein, transwell assays were used for investigating cell invasion and migration. Western blotting was performed to measure the expression of YAP. Results Ezrin was down-regulated after transfected with Ezrin siRNA in SGC-7901 ceils compared with NC (P〈0.05), the ability of invasion and migration significantly decreased (P〈0.05), expression of YAP was down-regulated observably (P〈0.05). Conclusion Down-regulation of Ezrin in SGC-7901 cells may suppress cells invasion and migration,probably via downregulating the YAP expression.
