摘  要:目的:利用槟榔碱饮水法初步构建与人口腔黏膜下纤维性变(OSF)发生发展相似的Balb/c小鼠OSF模型。方法:54只Balb/c小鼠随机分成实验组和对照组,每组27只。实验组采用500mg/L槟榔碱饮水法喂养8~16周后停药,对照组仅给予同等量自来水喂养。在实验第8周后,每隔2周随机处死两组小鼠各5只,进行颊部黏膜HE染色显微结构观察及Masson三色染色确定胶原堆积。结果:实验组在第12周后开始出现了不同程度的OSF样改变,表现为上皮萎缩、黏膜固有层、黏膜下层胶原堆积和微血管病变;病变进一步发展可波及深部肌层。对照组未见明显的组织形态学改变。结论:采用槟榔碱饮水法可快速诱发出小鼠颊部较典型、稳定的OSF模型。Objective: To establish a Balb/c mouse model induced by arecoline. The occurrence and development of OSF is similar to human OSF. Methods: 54 Balb/c mouse were assigned randomly into two groups: experimental group and control group. 500 mg/L arecoline was added into drinking water in experimental group for 8-16 weeks. The control group was given ordinary tap water. From Week 8, five mice from the experimental group and the control group respectively were killed every two weeks. Buccal mucosa were removed for histological assessment by HE, and collagen cumulate was verified by Masson staining. Results. From Week 12, the experimental group was observed to experience epithelium atrophy, collagen cumulated and micrangium pathologic changed in lamina propria and submucosa, further development of lesions could spread to the deep muscular layer. No abnormalities were found in the control group. Conclusion: Arecoline induced mouse buccal mucosa lesion successfully.
