丁苯酞注射液对蛛网膜下腔出血大鼠早期脑损伤的影响

摘  要:目的:观察丁苯酞(NBP)注射液对蛛网膜下腔出血(SAH)大鼠早期脑损伤(EBI)的影响,探讨其效果及可能机制。方法:建立SAH大鼠模型,将60只大鼠随机分为6组,分别为空白对照组(非手术组)、生理盐水对照组(NS对照组)、NBP组,每组又根据时点不同分为12h组与24h组,检测各组间不同时点丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、细胞间黏附分子-1(ICAM-1)以及细胞凋亡情况。结果 :NS对照组和NBP组的MDA、TNF-α、IL-1β、ICAM-1及细胞凋亡率均较非手术组升高(P<0.05),同一时点内(12h或者24h),NBP组MDA、TNF-α、IL-1β、ICAM-1及细胞凋亡率较NS对照组降低(P<0.05)。结论:在SAH后早期,MDA、TNF-α、IL-1β、ICAM-1的水平以及细胞凋亡率会升高,NBP在一定程度上能够减少这些炎性分子的表达,减轻炎性反应的发生,并降低SAH后细胞凋亡率,对SAH后的EBI起到一定的保护作用。Objective: To observe the affect of butylphthalide injection on early brain injury after subarach noid hemorrhage in rats, and explore its effects and possible mechanisms. Methods: Creating rat model of SAH,60 rats were divided into six groups randomly:blank control group, NS group, NBP group, and each group was divided into 12 hours group and 24 hours group. MDA,TNF-α,IL-β,ICAM-1 and condition of cell apoptosis were detected in each group in 12 hours and 24 hours. Results: The level of MDA,TNF -α,IL -β,ICAM-1 and condition of cell apoptosis all increased in NS group and NBP group than those in blank control group( P 〈0.05) ,in the same time(I2 hours or 24 hours) ,the level of MDA,TNF-α,IL-1β,ICAM- 1 and condition of cell apoptosis all decreased in NBP group than those in NS group. Conclusion: The level of MDA,TNF-α,IL-1β,ICAM-1 and cell apoptosis increase in the early stage of SAH,NBP can improve the condition of the early brain injury after the SAH through reducing the expression of MDA,TNF-α, IL-lβ, ICAM-1,easing the Inflammation and lowering the cell apoptosis to some extent.
