摘 要:目的研究证实,NF-κB在不同类型的肿瘤发生过程中均有异常活化,在肿瘤细胞的增殖过程中起重要作用。MIB2蛋白可以通过泛素化的途径降解IκB并最终实现对NF-κB的调控作用。本研究探讨MIB2蛋白在脑胶质瘤检出的意义,为临床上判断脑胶质的恶性程度提供参考。方法收集深圳市人民医院2013-01-01-2015-01-01手术切除并确诊为脑胶质的标本59例,采用免疫组化方法检测脑胶质瘤组织中MIB2蛋白的表达情况,并将MIB2与脑胶质瘤HE切片病理分级进行比较;以胶质瘤正常组织为对照组,采用蛋白质印迹法检测脑胶质瘤和正常脑组织MIB2、IκB和NF-κB表达。结果在脑胶质瘤组织中MIB2在低级别胶质瘤(low grade gliomas,LGG)Ⅰ和Ⅱ级组织中阳性表达率为57.89%(11/19),高级别胶质瘤(high grade gliomas,HGG)Ⅲ和Ⅳ级为85.00%(34/40),MIB2表达级别与胶质瘤的病理级别呈正相关,r^2=0.255,P<0.05。MIB2与脑胶质瘤HE切片病理分级显示,MIB2的表达与脑胶质瘤的恶性程度相关。MIB2、IκB和NF-κB的蛋白质印迹法结果表明,与正常脑组织相比,脑胶质瘤组织中MIB2和NF-κB含量显著升高,IκB含量显著降低。结论 MIB2在脑胶质瘤组织中表达显著升高,检测MIB2有助于判断脑胶质瘤的恶性程度。OBJECTIVE NF-κB signal pathway in gliomas is abnormal activation, and polyubiquitinated regulation was reported rarely. In this study, MIB2 in gliomas which regulated NF-κB signal pathway through polyubiquitinatedly degrading IκB and its clinical significance were explored. METHODS Totally 59 glioma samples in the Shenzhen People' s Hospital were collected from 2013-01-01 to 2015-01-01. The expression of MIB2 in 59 cases of glioma tissues in all grades were assayed by immunohistochemistry and compared with glioma pathology stained by HE. Western blot was used to detect the expression of MIB2, IκB and NF-~B in glioma tissue compared with its surrounding brain tissue. RESULTS The positive rates of MIB2 expression in low grade gliomas (LGG, Ⅰ and Ⅱ grade) and high grade gliomas (HGG, Ⅲ and Ⅳ grade) were 57.89 % (11/19) and 85.00 % (34/40) respectively. The difference was statistically significant between LGG and HGG, r2 = 8. 194, P〈0.05. The expression of MIB2 was closely positively relative to its pathology, r2 = 0. 255, P〈0.05. Compared with the pathological grading of gliomas stained by HE, MIB2 expression was associated with malig nant degrees of gliomas. Compared with surrounding brain tissue, the expression of MIB2 and NF- κB analyzed by Western blot increased significantly and the expression of IκB decreased significantly. CONCLUSIONS The expression of MIB2 in glioma tissue increases significantly. Detecting MIB2 in gliomas can be used for judging the malignancy degree.
