摘 要:为验证褐环粘盖牛肝菌95%乙醇提取物(Suillus luteus ethanol extract,SLE)抗糖尿病活性,本文采用抑制α-葡萄糖苷酶活性实验验证SLE的体外降血糖活性;建立高脂高糖饲料联合链脲佐菌素(STZ)诱导的糖尿病小鼠模型,检测SLE处理后模型小鼠血糖、血脂变化;组织切片法检测糖尿病小鼠病变胰脏、肝脏和肾脏的恢复情况。抑制α-葡萄糖苷酶活性检测结果表明,SLE的EC50值为3.269 mg/m L,表现出一定的体外降血糖活性。SLE处理后糖尿病模型小鼠血糖、三酰甘油(TG)、胆固醇(TC)降低,高密度脂蛋白胆固醇(HDL-C)升高,体重增加;SLE处理4周后,糖尿病模型小鼠的胰脏、肝脏、肾脏病变组织得到明显修复。本实验结果证明SLE具有较好的抗糖尿病活性,可用于相关保健品或药品的开发。In this study,ln vivo and in vitro treatments were carried out to investigate the hypoglycemie and antidiabetie effects of SuiUus luteus ethanol extract (SLE). SLE exhibited potent inhibitory activity against α-glueosidase in a dosedependent manner. The ECso of SLE was 3. 269 mg/mL. Streptozotoein-indueed diabetle mice showed that SLE can decrease the levels of blood glucose ,serum total cholesterol (TC), triglyeeride (TG), and low density lipoprotein (LDL- C) and increase the level of high density lipoprotein (HDL-C). The weight of the diabetic mice was increased after the SLE treatment. In addition, the pathological changes of pancreas,liver and kidney tissues in the mice model were recovered after the SLE was added. In conclusion, SLE exhibited better antidiabetic activity. It can be used for the development of health food or medicine.
