摘 要:目的:研究白藜芦醇(RES)对食管鳞癌Eca-109细胞侵袭迁移及上皮间质转化(EMT)的影响。方法:用不同浓度(50和100μmol/L)的RES处理Eca-109细胞,采用细胞划痕实验、Transwell小室实验检测细胞迁移侵袭能力,荧光定量PCR检测EMT蛋白E-cadherin、N-cadherin、Vimentin和MMP-2、MMP-9 mRNA的表达,Western blot检测E-cadherin、N-cadherin、Vimentin、MMP-2和MMP-9蛋白表达。结果:与对照组比较,RES各浓度(50μmol/L和100μmol/L)组均可明显抑制Eca-109细胞侵袭迁移(P〈0.01),下调N-cadherin、Vimentin、MMP-2、MMP-9 mRNA和蛋白表达(P〈0.05~P〈0.01),增加E-cadherin mRNA和蛋白表达(P〈0.01)。结论:RES抑制Eca-109细胞侵袭迁移能力,其机制可能通过下调MMP-2、MMP-9的表达,调控EMT蛋白(E-cadherin、N-cadherin和Vimentin)的表达、抑制上皮间质转化过程。Objective: To investigate of the effects of resveratrol( RES) on the invasion migration of Eca-109 cells and epithelialmesenchymal transition( EMT) in esophageal squamous cell carcinoma. Methods: The Eca-109 cells were treated with different dilutions( 50 and 100 μmol / L) of RES,then the wound-healing were performed. The cell migration and invasion was detected by transwell chamber assays. The mRNA and protein expressions of E-cadherin,N-cadherin,Vimentin,MMP-2 and MMP-9 were evaluated using real-time quantitative PCR and Western blot,respectively. Results: Compared with the control group,the other two RES treatment group( 50 and 100 μmol / L) could significantly inhibit the Eca-109 cell invasion and migration( P 0. 01),increase the mRNA and protein expressions of N-cadherin,Vimentin,MMP-2 and MMP-9( P 0. 05 to P 0. 01),and increase the mRNA and protein expression of E-cadherin( P 0. 01). Conclusions: Resveratrol can inhibit the invasion and migration of Eca-109 cells,which can be caused by down-regulating expressions of the MMP-2 and MMP-9,regulating the expressions of EMT protein( E-cadherin,N-cadherin and Vimentin),and inhibiting the epithelial mesenchymal transition process.
