小链霉菌HCCB10043中鸟氨酰脂类物质的鉴定及其生物合成基因的研究

摘  要:目的对首次从达托霉素产生菌小链霉菌(Streptomyces parvus)HCCB10043发酵液中分离得到的2个同系物,并进行结构鉴定,并研究其生物合成基因lyw A的功能。方法利用制备型液相色谱仪制备化合物1和化合物2,经质谱比对分析鉴定2个化合物的结构;采用同源重组方法敲除lyw A基因,并进行发酵验证。结果鉴定化合物1和化合物2为2个鸟氨酰脂类物质的前体,分别为N-α-3-hydroxy palmitoyl ornithine和N-α-3-hydroxy heptadecanealkanoyl ornithine;而缺失突变株的发酵液中不含化合物1和化合物2。结论 lyw A基因为小链霉菌中鸟氨酰脂类物质的合成基因。Objective To identify the structures of compound 1 and compound 2 that were firstly isolated from the fermentation broth of the daptomycin producing strain Streptomyces parvus HCCB10043 and to study the function of their biosynthetic gene lywA. Methods Compounds 1 and 2 were prepared by preparative liquid chromatography and their structures were identified by high resolutionmass spectrometry.Then the gene lywA was knocked out by homologous recombination and the result was verified by fermentation. Results Compounds 1 and 2 were identified as two precursors of omithine lipids, N-α- 3-hydroxy palmitoyl ornithine and N-a-3-hydroxy heptadecanealkanoyl ornithine respectively, and they were not found in the fermentation broth of the lywA deletion mutants. Conclusions The gene lywA is required for ornithine lipids biosynthesis in Streptomyces parvus.
