CD147、EGFR和HIF-1α在非小细胞肺癌中的表达及意义

摘　　要：目的检测基质金属蛋白酶诱导因子(CD147)、表皮生长因子受体(EGFR)和乏氧诱导因子1α(HIF-1α)在非小细胞肺癌(NSCLC)中的表达及临床意义。方法采用免疫组化SP法检测50例NSCLC与10例正常肺组织标本中CD147、EGFR和HIF-1α的表达,探讨其与肿瘤临床病理指标的关系。采用Western blot法检测NSCLC A549细胞系、SPC-A-1细胞系和正常肺上皮BEAS-2B细胞系中CD147、EGFR和HIF-1α的表达,分析其临床意义。结果CD147、EGFR和HIF-1α在NSCLC组织中的阳性表达率分别为76%、78%和84%,高于正常肺组织中的10%、40%和30%(P<0.05);且与肿瘤的分化程度及有无淋巴结转移相关,CD147还与TNM临床分期有关(P<0.05)。CD147、EGFR和HIF-1α在NSCLC中的表达两两之间均存在正相关(P<0.05);在NSCLC A549和SPC-A-1细胞系中的相对表达量均高于正常肺上皮BEAS-2B细胞系(P<0.05);且相对表达量在三种细胞系之间的差异均有统计学意义(P<0.05)。结论 CD147、EGFR和HIF-1α在NSCLC的发展、侵袭及转移过程中起重要作用,可能也是NSCLC的临床治疗靶点。Objective To investigate the expressions and significance of extracellar matrix metallo proteinase inducer（CD147）,epidermal growth factor recepter（EGFR）and hypoxia inducible factor-1α（HIF-1α）in non-small cell lung cancer（NSCLC）.Methods The expressions of CD147,EGFR and HIF-1αwere detected by immunohistochemical SP staning in 50 samples of NSCLC and 10 samples of normal lung tissues.The relationship of the expressions of them with clinicopathological indicators were analyzed.Western blot method was used to detect the expressions of CD147,EGFR and HIF-1αin NSCLC A549,SPCA-1cell line and normal lung epithelial BEAS-2Bcell line.The clinical significance of the expressions of them was analyzed.Results The positive expression rates of CD147,EGFR and HIF-1αwere 76%,78% and 84%,respectively,which were significantly higher than 10%,40% and 30%in normal lung tissues.The expressions of CD147,EGFR and HIF-1αwere closely associated with tumor differentiation and lymphatic metastasis of NSCLC（P〈0.05）.Meanwhile the expression of CD147 was closely associated with TNM clinical stages of NSCLC as well（P〈0.05）.There were significant positive correlations among the expressions of CD147,EGFR and HIF-1αin NSCLC（P〈0.05）.Compared with normal lung epithelial BEAS-2Bcell line,CD147,EGFR and HIF-1αwere highly expressed in NSCLC A549,SPCA-1cell line（P〈0.05）.There were significant positive correlations among the expressions of CD147,EGFR and HIF-1αin the three cell lines（P〈0.05）.Conclusion CD147,EGFR and HIF-1αplay an important role in the development,invasion and metastasis of NSCLC and may be the targets for the treatment of NSCLC.
