摘 要:目的本研究将FTIR技术用于大鼠酒精性肝病理模型检测,通过谱图能反映出肝细胞酒精性损害的微小变化,FTIR技术快速、精确、简便,可以在分子水平为酒精性肝病的早期诊断提供依据,为临床研制开发酒精性肝病新药及疗效判定提供基础理论依据。方法本实验通过90只雌雄性Wistar大鼠灌胃40%浓度酒精9ml/kg体重,获取60例大鼠酒精性肝损害组织和30例正常肝组织标本,应用FTIR光谱仪进行扫描。对获得的酒精性肝损害组织图谱应用Spapro2软件分析,测定参数包括组织图谱峰强、峰位、半峰宽,并计算峰强比值。结果正常肝组织和酒精性肝损伤组织间红外光谱有统计学差异,光谱技术检测的酒精性肝组织同病理学诊断金标准的结果进行比较,此判别方法的灵敏度和特异度为96.7%。结论 FTIR光谱技术可以达到快速准确的鉴别酒精性肝损害的目的。Objectives This research will FTIR technology used in rats with alcoholic liver disease model checking, through the spectrum diagram can reflect the subtle change of alcoholic liver cell damage, FTIR technology is fast, accurate, convenient, can at the molecular level to provide the basis for early diagnosis of ALD, for clinical research and development of ALD drugs provide theo- ry basis. Methods Using the ethanol lavage method. This experiment through 90 female male Wistar rats by gastric 40% concen- tration alcohol 9 ml/Kg weight,for 60 cases of alcoholic liver injury in rats and 30 cases of normal liver tissue samples, using an FT- IR spectrometer. Results Compare two groups of various pa- rameters, to test for normal distribution and non-normal distribution line Mann. Calculate sensitivity and specificity96. 7% , discriminant classification results with the pathological gold standard for comparison FTIR. Conclusions FTIR spectroscopy can achieve rapid, accurate identification of the purpose of the alcoholic liver damage.
