四季三黄丸高效液相指纹图谱与其体外抗氧化活性的灰关联度分析  被引量:2

摘  要:目的:建立中药制剂四季三黄丸HPLC指纹图谱,确定10批四季三黄丸指纹图谱共有峰与其体外抗氧化活性作用贡献大小。方法:利用HPLC研究四季三黄丸指纹图谱,利用DPPH自由基清除实验研究其体外抗氧化作用,利用灰关联度分析法研究其谱效关系。结果:将10批经过积分处理的四季三黄丸样品的色谱图导入国家药典委员会《中药色谱指纹图谱相似度评价系统》2004A版软件,确定了33个共有峰,批次间相似度均在0.9以上,通过与对照品的保留时间和在线紫外光谱进行比较,对其中9个特征峰进行了定性;四季三黄丸体外抗氧化作用是多化学成分共同作用的结果,通过实验确定了各特征峰对DPPH自由基清除作用贡献大小顺序,其中11号峰和31号峰贡献最大,4号峰和13号峰贡献最小。结论:本实验所构建的四季三黄丸HPLC指纹图谱方法简单,重复性好,可为四季三黄丸的鉴别和质量评价提供参考,四季三黄丸HPLC指纹图谱与其体外抗氧化活性之间有一定的相关性。OBJECTIVE To establish HPLC fingerprint of Siji Sanhuang Pills (SJSHW) and determine contribution of common peaks of the fingerprint to the antioxidant activity in vitro, METHODS HPLC was used for development of fingerprint of 10 batches of SJSHW samples. Evaluation of their antioxidant activities in vitro were performed by DPPH free radical scaven- ging experiment and finally the gray relational analysis was used for studying relationship of fingerprint and antioxidant effects. RESULTS Chromatograms of 10 batches of Siji Shanhuang Pill samples after integral processing were introduced into the soft- ware of "Traditional Chinese Medicine (TCM) Chromatographic Fingerprint Similarity Evaluation System" (2004A version) promulgated by National Pharmacopoeia Committee. A total of 33 common peaks were determined and the similarities between different batches were all more than 0. 9. Compared with the retention time and online UV spectra of reference substance, nine characteristic peaks were identified. The antioxidant experiment proved that its antioxidant activity in vitro was the result of combined effects of chemical components, and confirmed the order of contributions of characteristic peaks to DPPH free radical scavenging effects. Peak 11 and 31 had the maximal contribution, and peak 4 and 13 had the minimal. CONCLUSION The method for establishing HPLC fingerprint of Siji Sanhuang Pills is simple, repeatable, can provide a reference for identification and quality evaluation of SJSHW. There is a certain correlation between the fingerprint of SJSHW and its antioxidant activity in vitro.
