摘 要:目的:探讨抑制过氧化物酶1(Prx-1)对肝癌细胞放射敏感性的影响。方法:选用人肝癌Hep G2细胞,用分次放疗放射剂量递增法诱导放射抵抗的肝癌细胞(RR-Hep G2),检测RR-Hep G2细胞与亲代Hep G2细胞中Prx-1的表达,以及两种细胞在接受相同放射处理后存活率与迁移、侵袭能力。用表达sh RNA-Prx-1的质粒转染两种细胞后,再次检测上述指标的变化。结果:与亲代Hep G2细胞比较,RR-Hep G2细胞Prx-1的m RNA与蛋白表达明显增高;放射处理后,RR-Hep G2细胞的存活率升高,迁移能力及侵袭能力增强(均P<0.05)。转染sh RNA-Prx-1后,与各自转染阴性对照序列的细胞比较,两种细胞Prx-1的m RNA与蛋白表达均明显降低;接受放射处理后,两种细胞的存活率均降低,迁移能力以及侵袭能力均明显减弱(均P<0.05)。结论:肝癌细胞对放射的敏感性降低可能与Prx-1的表达有关,调控Prx-1的表达可望成为增强肝癌细胞放射敏感性的有效途径。Objective: To investigate the ef ect of peroxiredoxin 1(Prx-1) inhibition on radiosensitivity of hepatocellular carcinoma(HCC) cells. Methods: Using human HCC Hep G2 cells, the radioresistant HCC cells(RR-Hep G2) were induced by fractionated irradiation with ascending radiation dose levels. In RR-Hep G2 cells and their parental Hep G2 cells, the Prx-1 expression was examined, and at er undergoing the same radiation treatment, the survival rate and the invasion and migration abilities were determined. Then, in these two types of cells after transfection with the plasmids bearing shR NA-Prx-1, the above parameters were determined again.Results: In RR-Hep G2 cells compared with their parental Hep G2 cells, both Prx-1 m RNA and protein expressions were signii cantly increased; the survival rate, and invasion and migration abilities were signii cantly increased at er radiation treatment(all P〈0.05). At er sh RNA-Prx-1 transfection, in both types of cells comparedwith their counterparts transfected with negative control sequences, both Prx-1 m RNA and protein expressions were signii cantly decreased; the survival rate, and invasion and migration abilities were signii cantly decreased at er radiation treatment(all P〈0.05). Conclusion: The decreased radiosensitivity in HCC cells may probably be associated with Prx-1 expression, and regulating its expression could potentially be an effective approach for enhancing radiosensitivity of HCC cells.
