摘 要:目的 探讨中国汉族人群中白介素33(IL-33,interleukin-33)基因单核苷酸多态性(SNP,single nucleotide polymorphism)与脓毒症发生风险及疾病严重程度的相关性。方法 采用病例-对照研究设计,标签SNP(tagging SNP,tagSNP)策略,应用Sequenom MassARRAY?SNP检测技术完成对IL-33基因的标签SNPs分型。采用非条件Logistic 回归分析,并对性别、年龄、吸烟和饮酒、APACHEⅡ评分、慢性病状态等混杂因素进行校正后,评估IL-33单核苷酸多态性与脓毒症易感性的关系,以及脓毒症继发的严重脓毒症、脓毒症休克、死亡等表型的相关性。 结果 在所选的7个IL-33标签SNPs(rs10118795 C/T,rs16924159 G/A,rs1891385 A/C,rs2210463 A/G,rs72614079 A/G,rs76864631 G/A,rs7849201 C/T)中,rs76864631不符合Hardy-Weinberg平衡定律,其余6个多态位点及单体型与脓毒症的发生风险和疾病的严重程度均无明显相关性。 结论 IL-33基因多态性可能与中国汉族人群脓毒症的发生、发展和病程转归无关。Objective To explore the association between interleukin-33 polymorphisms and the susceptibility to and severity sepsis in Chinese Han. Methods A case-control study was conducted. The tagSNPs were selected from the interleukin-33 gene by linkage disequilibrium structure and the genotypes of all samples were determined by Sequenom MassARRY system. The associations with susceptibility to and phonotypes for severity of sepsis ( including severe sepsis, sepsis shock and death, etc. ) were estimated by non conditional logistic regression adjusted by age, sex, smoking, drinking, chronic disease status, APACHE Ⅱ score and primary disease status. Results Seven tagSNPs (rs10118795 C/T, rs16924159 G/A, rs1891385 A/C, rs2210463 A/G, rs72614079 A/G, rs76864631 G/A, rs7849201 C/T) were used for final genotyping analysis. Except for rs76864631, genotype frequencies of our patients and controls both confirmed to the Hardy- Weinberg equilibrium. There was no significant association between the 6 tagSNPs and susceptibility and disease severity of sepsis, after correction for muhiple comparisons ( P 〉 0. 0 125 ). Conclusions The interleukin-33 gene polymorphisms might have not a correlation with the susceptibility to and severity of sepsis in Han Chinese population.
