电针下合穴对十二指肠溃疡模型大鼠组织高迁移率族蛋白1及核因子-κB等的影响  被引量:1

摘  要:目的:对比观察电针十二指肠溃疡模型大鼠下巨虚、足三里、上巨虚、阳陵泉等穴后对十二指肠组织肿瘤坏死因子-α(TNF-α)、高迁移率族蛋白B1(HMGB1)及核因子-κB(NF-κB)表达的影响,探讨小肠下合穴下巨虚治疗对应腑病是否存在相对特异性。方法:将70只健康SD大鼠随机分为空白、模型、下巨虚、足三里、上巨虚、阳陵泉、承筋组共7组,每组10只,雌雄各半。除空白组外均皮下注射盐酸半胱胺建立十二指肠溃疡大鼠模型,造模成功并治疗10 d后,肉眼观察大鼠十二指肠黏膜溃疡情况,ELISA法检测十二指肠组织TNF-α、HMGB1含量,免疫组化法检测十二指肠中NF-κB的表达。结果:1与模型组比:5个穴位组TNF-α、HMGB1含量均降低(P<0.01),且上巨虚、下巨虚和足三里组溃疡评分及NF-κB的表达依次降低(P<0.05或P<0.01);2与下巨虚组比:阳陵泉组和承筋组评分、TNF-α、HMGB1含量以及NF-κB的表达均偏高(P<0.01或P<0.05),上巨虚组TNF-α、HMGB1含量亦增高(P<0.05),足三里组较之差异均无统计学意义(P>0.05)。结论:1电针下巨虚对DU大鼠炎症干预效果明显优于阳陵泉、承筋穴及上巨虚穴,提示下巨虚穴对DU具有一定的相对特异性;2足三里穴效果与下巨虚穴相当,二穴靶器官效应可能存在交集,其具体机制和内涵仍有待进一步研究。Objective: To explore whether the lower He points have relative specificity when treating its corresponding viscera disease and to observe the expression level of duodenal tissue TNF-α,high mobility protein B1 and Nuclear Factor-κB in duodenal ulcer model rats by electroacupuncture at Xiajuxu( ST39),Zusanli( ST36),Shangjuxu( ST37),Yanglingquan( GB34) and Chengjin( BL56). Methods: Seventy SD rats were randomly and divided into 7 groups: blank group,model group,Xiajuxu( ST39)group,Zusanli( ST36) group,Shangjuxu( ST37) group,Yanglingquan( GB34) group and Chengjin( BL56) group. Set up the Duodenal ulcer rat models through injection of cysteamine hydrochloride. And then the five treatment groups respectively received electroacupuncture on its corresponding points for 10 days. After treatment,duodenal tissues were collected to make DU score by eye first. ELISA was used to check models' duodenal tissue TNF-α,HMGB1 and NF-κB levels. Results: Compared with the model group,ulcer lesions of Zusanli( ST36) group,Shangjuxu( ST37) group,Xiajuxu( ST39) group were improved,the ulcer score,duodenal tissue TNF-α,HMGB1 and NF-κB levels were significantly decreased( P 0. 01). The intervention effect of Zusanli( ST36) group and Xiajuxu( ST39) group was better than that of the Yanglingquan( GB34) and Chengjin( BL56) group( P 0. 01 or P 0. 05). Conclusion: The effect of intervention DU inflammation in rats when EA at Xiajuxu( ST39) points was better than Yanglingquan( GB34) and Chengjin( BL56),slightly better than Shangjuxu( ST37),which proves that Xiajuxu( ST39) has relative specificity. Besides,the anti-inflammatory effects of Zusanli( ST36) was roughly equal to Xiajuxu( ST39),which might be concerned to overlap parts of target organs effect. Its mechanism still needs further research.
