扶正法与祛邪法对肺癌A549细胞凋亡的影响及其机制研究

摘　　要：目的:研究扶正法和祛邪法对肺癌A549细胞凋亡的影响并探讨其作用机制。方法:常规培养A549细胞,按实验分组干预,MTT法观察A549细胞增殖情况,流式细胞仪观察A549细胞凋亡情况,western blot法检测凋亡相关因子BCL-2、Caspase-3和Survivin的表达情况。结果:芪贞固本方、芩莲正积方对A549细胞增殖均有一定程度的抑制作用,且呈浓度和时间依赖性,均能显著提高A549细胞早期凋亡率,上调Caspase-3的表达,下调BCL-2、Survivin蛋白的表达,与对照组相比有显著差异性(P<0.05)。结论:扶正法和祛邪法用于肺癌治疗可诱导肺癌细胞凋亡,其机制可能为上调Caspase-3的表达,下调BCL-2、Survivin蛋白的表达。Objective： To study the effect of strengthening healthy Qi and eliminating pathogenic factors on the apoptosis of lung cancer line of A549 and its mechanism. Methods： To cultivate the A549 cell line and intervene the cell according to the groups.MTT was performed to reveal the effect on cell proliferation and flow cytometry was used to determine the apoptosis. And the expressions of BCL-2,Caspase-3 and Survivin were detected by Western blot. Results： Compared to the control group,there was a time-and dose-dependent inhibition on cell proliferation of A549 by Qizhen Guben Formula and Qinlian Zhengji Formula. And they all can increase the early apoptosis rate,promote the expression of Caspase-3 and down-regulate the expressions of BCL-2 and Survivin significantly（ P〈 0. 05）. Conclusion： Strengthening healthy Qi and eliminating pathogenic factors can induce the apoptosis of lung cancer cell. The mechanism may be related to promoting the expression of Caspase-3 and down-regulating the expressions of BCL-2 and Survivin.
