摘　　要：目的:探讨在肾癌细胞系中,沉默PDCD4对786-0细胞增殖能力的影响。方法:将786-0细胞分为两组(PDCD4 siRNA和阴性对照siRNA),分别转染PDCD4 siRNA和negative control siRNA,采用EDU增殖实验研究沉默PDCD4对肿瘤细胞增殖的影响。结果:EDU增殖实验中,阴性对照组(NC组)的增殖率为(28.6±2.3)%,PDCD4siRNA组的增殖率为(44.7±3.6)%,两组差异有统计学意义(P<0.05)。结论:沉默PDCD4后786-0细胞的增殖能力明显比阴性对照组强,表明PDCD4具有促进肾癌细胞增殖的作用。Objective The aim of this study was to study the effects of silencing PDCD4 on proliferation ability of 786- 0 cells in renal cancer cell line. Method 786- 0 were assigned to two groups（ negative control group,PDCD4 siRNA group）. we transfected the 786- 0 cells with PDCD4 siRNA and the negative control siRNA,and investigated the effects of silencing PDCD4 on tumor cell proliferation with EDU proliferation assay. Results In EDU proliferation assay,the percentage of new cells which incorporated EDU was（ 28. 6 ± 2. 3） % in the negative control siRNA group and（ 44. 7 ± 3. 6） % in PDCD4 siRNA transfected cells,the difference was statistically significant（ P〈0. 05）. Conclusion After silencing PDCD4,compared to the negative control group,the tumor cell proliferation of 786- 0 cells in the PDCD4 siRNA group was significantly lower,it is suggested that PDCD4 promotes cell proliferation in renal cancer cell.
