厄贝沙坦对乳腺癌MCF-7细胞的增殖抑制作用及对VEGF表达的影响

摘　　要：目的研究厄贝沙坦对乳腺癌MCF-7细胞增殖抑制作用及对血管表皮生长因子(VEGF)表达水平的影响。方法采用噻唑蓝(MTT)法检测厄贝沙坦对乳腺癌MCF-7细胞分别作用24 h、48 h、72 h后的抑制率,用酶联免疫吸附法(ELISA)检测厄贝沙坦高、中、低三个剂量(7.2μg/ml、3.6μg/ml、1.8μg/ml)对乳腺癌MCF-7细胞干预前后,乳腺癌MCF-7细胞中VEGF表达水平的差异,以顺铂作为阳性对照组(给药浓度为5μg/ml)。结果厄贝沙坦对乳腺癌MCF-7细胞的半数抑制浓度(IC50)为2.3μg/ml;厄贝沙坦对细胞增殖有显著抑制作用,且随着浓度的增加而增强,三个剂量组与空白对照组比较差异有统计学意义(P<0.05或0.01);厄贝沙坦作用组中VEGF的表达水平与空白对照组比较差异有统计学意义(P<0.01),厄贝沙坦高剂量组和顺铂阳性对照组VEGF的表达差异无统计学意义(P>0.05)。结论厄贝沙坦对乳腺癌MCF-7细胞的增殖具有显著抑制作用,可以下调血管表皮生长因子的表达水平。Objective To investigate the effects of Irbesartan on expression level of VEGF（ vascular epidermal growth factor） and its inhibition effect on the proliferation of MCF-7 in breast cancer. Methods The MTT method was used to detect the inhibition rate of irbesartan on the MCF-7 cells after effecting 24 h,48 h and 72 h,respectively. Enzyme-linked immunoabsorbent assayed（ ELISA） was used to detect the level of VEGF in MCF-7 cells influenced by the combination use of irbesartan（ the dose of irbesartan were divided into high-dose,middle-dose and low-dose which was 7. 2 μg / ml,3. 6 μg / ml and 1. 8 μg / ml,respectively）. Cis-platinum was taken as positive control group（ concentration of drug was 5 μg / ml）. Results The 50% inhibition concentration（ IC50） of irbesartan on MCF-7 was 2. 3 μg/ml. Irbesartan can inhibit the MCF-7cancer cells significantly,the inhibiting effect was enhanced while increasing the inhibitor＇s dosage,difference between the three dosage groups and the blank control group was statistically significant（ P〈0. 05 or 0. 01）. Difference of expression levels of VEGF between the irbesartan effecting groups and blank control group was statistically significant（ P〈0. 01）,but difference between high-dose group and cis-platinum positive control group was not statistically significant（ P〉0. 05）. Conclusion Irbesartan can inhibit the proliferation of MCF-7 cancer cells in breast cancer significantly,and lower expression levels of VEGF.
