摘 要:目的考察间歇吸氧对小鼠肺、脑组织中抗氧化蛋白Peroxiredoxin 6(Prx 6)含量的影响,探讨间歇吸氧对抗氧中毒的机制。方法将小鼠随机分为正常组、持续暴露后即刻组、间歇暴露后即刻组、持续暴露后24 h组和间歇暴露后24 h组,通过不同暴露方法,分别取小鼠肺、脑组织采用免疫组织化学和Wester blot等方法测定Prx 6的含量。结果经高压氧暴露的各组小鼠肺组织中Prx 6的含量均明显高于正常组(P<0.05或P<0.01),持续暴露后即刻组与间歇暴露后即刻组差异无统计学意义(P>0.05),间歇暴露后24 h组含量明显高于持续暴露后24 h组(P<0.01),并且高于各自对应的暴露后即刻检查的结果(P<0.01)。经高压氧暴露的各组小鼠脑组织中Prx 6的含量均明显低于正常组(P<0.01),持续暴露后即刻组与间歇暴露后即刻组差异无统计学意义(P>0.05),间歇暴露后24 h组与持续暴露后24 h组高于各自对应的暴露后即刻检查的结果(P<0.05),且间歇暴露后24 h组含量接近正常组(P>0.05)。结论间歇吸氧可以促进肺、脑组织中Prx 6的含量增加,从而增强组织的抗氧化能力。Objective To investigate the effects of intermittent hyperbaric oxygen exposure on the contents of peroxiredoxin 6 (Prx 6) in the lung and brain of mice, and to explore the mechanism underlying intermittent hyperbaric oxygen exposure protecting against Oxygen toxicity. Methods Mice were randomly divided into normal group, immediate group after continuous exposure, immediate group after intermittent exposure, live 24 h group after continuous exposure, live 24 h group after intermittent exposure. The content of Prx 6 in the lung and brain were detected using immunohistochemistry and western blot analysis respectively. Results In lung, compared with normal group, Prx 6 contents of all mice experiencing hyperbaric oxygen exposure were increased (P 〈 0,05 or P 〈 0.01). There was no significant difference between immediate group after continuous exposure and immediate group after intermittent exposure (P 〉 0.05). The Prx 6 content of live 24 h group after intermittent exposure was significantly higher than that of live 24 h group after continuous exposure (P 〈 0.01), and the result in each live 24 h group was also higher than that in each immediate group (P 〈 0.01). In brain, compared with normal group, Prx 6 contents of all mice experiencing hyperbaric oxygen exposure were decreased (P 〈 0.01). There was no significant difference between immediate group after continuous exposure and immediate group after intermittent exposure (P 〉 0.05). The Prx 6 contents in each live 24 h group was higher than that in each immediate group (P 〈 0.05), and the content of live 24 h group after intermittent exposure was close to the normal group (P 〉 0.05). Conclusion Intermittent hyperbaric oxygen exposure can facilitate the expression of Prx 6 in lung and brain, and this may be related to the enhanced ability to protect against oxygen toxicity.
