香烟烟雾对青春期雄性大鼠GnRH、IGF-1、LH、FSH及T的影响

摘　　要：目的研究香烟烟雾对青春期雄性大鼠下丘脑中促性腺激素释放激素(GnRH)、血清中胰岛素样生长因子-1(IGF-1)、促黄体激素(LH)、卵泡刺激素(FSH)、睾酮(T)的影响。方法选用5周龄SD大鼠80只,分为对照组与吸烟组,吸烟组采用呼吸道静式染毒,每天1次,每次2h,连续8w;实验第2周、第4周、第6周与第8周分别处死对照组与吸烟组大鼠各10只,放射免疫法测定血清中睾酮、黄体生成激素、卵泡刺激素及下丘脑促性腺激素释放激素,ELISA法测定血清中IGF-1含量。结果吸烟组大鼠血清中T、LH含量自第4周起明显低于对照组(P<0.05);吸烟组大鼠血清T、LH含量随染毒时间延长明显下降(P<0.05);对照组与吸烟组大鼠血清中FSH含量稳定,自第2周起吸烟组FSH含量明显低于对照组(P<0.05);对照组与吸烟组大鼠下丘脑GnRH含量均随时间延长而升高,吸烟组与对照组比较差异无统计学意义(P>0.05);大鼠血清中IGF-1含量随时间的延长而逐渐下降,吸烟组大鼠血清中IGF-1在第8周含量明显低于第2周、第4周与第6周(P<0.05),吸烟组第2周、第6周和第8周IGF-1均明显低于对照组(P<0.05)。结论香烟烟雾是青春期雄性大鼠生殖系统发育的有害因素,对青春期雄性大鼠的下丘脑-垂体-睾丸轴产生明显的损害作用。Objective To study effects of cigarette smoke on the gonadotropin-releasing hormone（GnRH）of hypothalamus,and luteinizing hormone（LH）,follicle stimulating hormone（FSH）,testosterone（T）,and insulin-like growth factor-1（IGF-1）in pubertal male rat serum.Methods 80 SD rats at the age of 5weeks were randomly divided into control group,and smoking groups,which was inhaled cigarette smoke for 2hper day and continued for 8weeks.By the end of the 2nd,4th,6th,8th week,10 male mice in the smoking group and the control group were respectively executed to observe GnRH in hypothalamus and LH,FSH and T in serum determined by radioimmunoassay kit,and IGF-1in serum by ELISA.Results GnRH in either the control group or the smoking group increased with time going,but there was no statistical significance between them（P〈0.05）;T and LH in the smoking group were significantly lower than that in the control group after the 4th weeks（P〈0.05）,and they were reduced with the extension of exposed time（P〈0.05）.FSH was stable in the control group and the smoking group,and it was distinctly lowered since the 2nd week in the smoking group than in the control group（P〈0.05）;IGF-1levels decreased with time going.IGF-1in smoking group at the 2nd week,the 6th week and the 8th week was significantly lower than those in the control group（P〈0.05）,and at the 8th week it was significantly lower than that at the 2nd week,the 4th week and the 6th week（P〈0.05）.Conclusion Cigarette smoke is the harmful factor in developing of pubertal rat reproductive system,and negatively affect on the Hypothalamic-pituitary-Testicular axis hormones.
