摘 要:目的了解衰老背景以及衰老背景下小鼠p21功能缺失胃窦细胞线粒体形态改变,探讨衰老背景下小鼠p21功能缺失胃癌易感的发生机制。方法小鼠端粒酶和wrn基因同时敲除(mTerc-/-wrn-/-)并在后代(G4)表现稳定的衰老表型,取同代同龄的野生型(WT)、衰老以及衰老背景下p21功能缺失的雄鼠各15只,获得胃窦组织进行透射电子显微镜观察细胞线粒体形态。结果 WT、衰老以及衰老背景下p21功能缺失的小鼠胃窦细胞线粒体异常率分别是13.3%、53.3%和93.3%。结论衰老背景下p21的功能缺失导致胃窦细胞线粒体发生明显的形态异常,由此可能促进癌变的风险。Objective In order to know about the change of mitochondria in pyloric antrum cells under mice of aged model and p21 loss of function in aged mice. To investigate the mechanism of gastric carcinoma in aged mice which lost of p21. Methods Telomerase deficiency and wrn null(m Terc-/-wrn-/-) mice were generated,which in later generation(G4) showed out stable type of aged. To take 15 mice from WT, aged and p21 loss of function in aged, respectively. To watch the shape of mitochondria in pyloric antrum cells by transmission electron microscope.Results The rates of abnormal mitochondria in pyloric antrum cells from WT, aged and p21 loss of function in aged were 13.3%, 53.3%and 93.3% respectively.Conclusion The obviously abnormal of mitochondria configuration in pyloric antrum cells which belongs to p21 loss of function in aged mice. This phenomenon may promote the risk of gastric carcinoma.
