摘 要:目的观察复合型寡聚糖(CTO)和单一寡糖(COS)对雄性小鼠生殖能力的影响及作用途径。方法选择21日龄雌、雄性小鼠72只,随机分为NC组、COS组和CTO组(COS:FOS:YP=1:1:1),每组24只,雌、雄各半分别饲养。实验开始后7w龄时同组雌、雄随机1:1合笼5d。待仔鼠出生后3、6、12w龄时分别采集相关样品,观察了血清睾酮(T)、促卵泡素(FSH)、促黄体生成素(LH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、精子活力、DDX4蛋白表达及睾丸组织结构等指标。结果 (1)实验6w龄时睾丸系数CTO和COS组比NC组均提高,但CTO组差异显著(P<0.05)。12w龄时睾丸系数CTO、COS组比NC组均增加,组间差异显著(P<0.05);(2)12w龄时附睾精子存活率CTO和COS组均比NC组显著提高(P<0.05)。a级精子百分率CTO、COS组均优于NC组(P<0.05);(3)12w龄时曲细精管生精细胞和成熟精子数量CTO和COS组均比NC组提高(P<0.05);(4)3、6、12w龄时血清T含量CTO组最高,其次COS组,NC组最低,组间差异显著(P<0.05);(5)睾丸SOD、GSH-Px含量,3、6、12 w龄CTO组最高,其次COS组和NC组,而MDA含量呈相反的趋势,各组间差异显著(P<0.05);(6)3、6、12w龄时睾丸中DDX4蛋白表达量CTO组比COS、NC组均有提高,其中3、12w龄CTO和NC组差异显著(P<0.05)。结论 CTO优于单一COS可以提高雄性小鼠的生殖能力,并可提高睾丸中DDX4蛋白的表达量。Objective To investigate the effects of chitosan oligosaccharide(COS) and complex oligosaccharide(CTO) on male reproductive capacity in mice and the mechanisms involved. Methods Seventy-two adult mice(3-week-old) were randomly assigned to 3 groups: control group(NC), COS-treated group and COT-treated group. At 7 weeks old, each male mouse was mated with a female for 5 days to obtain the F1-generation. Key reproductive-related parameters of the F1-generation, including levels of T, FSH, LH, SOD, GSH-Px and MDA, sperm activities, DDX4 protein expression and testis index, were analyzed at the age of 3, 6, and 12 weeks, respectively. Results(1) Testis index were increased in 6-week old COS and COT-treated mice as compared to the NC mice, but only the difference between the CTO-treated mice and the NC were statistically significant.(2) Sperm viability and a-grade sperm percentages were significantly increased in the 12-week old CTO-treated mice as compared to the control group(P0.05).(3) Numbers of spermatogenic cells and mature sperm in the seminiferous tubules were significantly increased in the 12-week CTO-treated mice(P0.05).(4) Serum testosterone concentrations were significantly increased at all ages of the COS and CTO-treated mice as compared to the control group(P0.05), and the CTO-treated mice manifested the highest levels of serum testosterone.(5) Although both CTO and COS-treated mice showed increased levels of DDX4 in testis, only the difference between the 3 and 12-week old CTO-treated group and the control group was statistically significant(P0.05). Conclusion Complex oligosaccharide could increase the reproductive capacity via enhancing the expression of testicular DDX4 in male mice.
