摘 要:目的:观察复方土茯苓颗粒对高尿酸血症大鼠血尿酸(UA),黄嘌呤氢化酶(XO)活性及肝脏中黄嘌呤氧化酶(XDH)mRNA表达水平的抑制作用。方法:48只SD大鼠随机分成正常组,模型组,非布司他组(4 mg·kg^(-1)),复方土茯苓颗粒低、中、高剂量组(1,2,4 g·kg^(-1)),除正常组外,其余各组制备高尿酸血症模型及给予相应药物干预,每天1次,共给药5 d。实验结束后测定各组大鼠的血UA,肝XO活性以及肝中的XDH mRNA表达水平。结果:模型组大鼠的血UA,肝XO活性,肝XDH mRNA表达水平均比正常组明显升高(P<0.05);经药物治疗后,各给药组大鼠血UA,肝XO活性,肝XDH mRNA表达水平均比模型组明显降低(P<0.05),复方土茯苓颗粒低剂量组与模型组无明显差别。结论:复方土茯苓颗粒可以有效降低高尿酸血症大鼠血UA,并可以抑制XO活性与肝脏中XDH mRNA表达水平。Objective: To study the inhibition effect of compound Tufuling granules (CTG) on uric acid (UA), activity of xanthine oxidase (XO), and xanthine dehydrogenase(XDH) mRNA in hyperuricemic rats. Method: The 48 SD rats were randomly divided into normal group, model group, febuxostat group (4 mg · kg^-1), CTG low-dose group, CTG mid-dose group and CTG high-dose group (1, 2, 4 g · kg^-1). Rats in all other groups except normal group were used to establish hyperuricemia (HUA) models and received corresponding drugs for intervention, once a day for 5 days. After experiment, UA of serum, activity of XO, and XDH mRNA in liver were detected. Result: In model group, levels of UA, activity of XO, and XDH mRNA expression levels in liver were significantly higher than normal group (P〈0.05). After treatment, levels of UA, activity of XO, and XDH mRNA expression levels were significantly lower in various treatment groups than those in model group (P〈0.05), but there was no significant difference between CTG low dose group and model group. Conclusion: CTG may reduce the UA of serum, and inhibit the activity of XO and XDH mRNA expression level in hyperuricemic rats.
