摘 要:目的探讨P53联合紫杉醇在宫颈癌中的抑制作用。方法利用宫颈癌细胞系He La细胞进行实验研究,利用MTT实验对单独加入P53重组蛋白、紫杉醇和P53联合紫杉醇对细胞增殖进行检测,两者均不加为对照组,同样的设计思路对细胞单克隆进行检测,最后用western-blot进行4种细胞蛋白的检测。结果重组蛋白联合紫杉醇时抑制宫颈癌细胞的生长,且抑制作用具有统计学差异;细胞单克隆形成数目也是减少,在初步探讨其作用机制时发现VEGF表达减少。结论 P53重组蛋白联合紫杉醇能有效抑制宫颈癌细胞生长,其潜在作用机制是抑制VEGF的表达。Objective To detect the inhibitory effect of recombinant human P53 combined with paclitaxel on human cervical cancer cell and its mechanism. Methods Human HeLa cell was used to detect the cell proliferation,and add P53 or paclitaxel alone, and both of them as experiment, blank as a control, the congenic assay was detected, and the protein was detected by western-blot. Results The growth of HeLa cells decreased after add P53 and paclitaxel, the P53 and paclitaxel alone also decreased, P53 and paclitaxel had statistical difference,P 〈 0. 001, and the ability of congenic decreased after add P53 and paclitaxel, and the expression of VEGF decreased. Conclusion The inhibitory ability of recombinant human P53 combined with paclitaxel on human cervical are efficient,its potential mechanism of action is inhibition of the expression of VEGF.
