摘  要:目的研究粗壮女贞不同萃取部位总黄酮和粗多糖含量差异,分析该差异与其抑菌活性差异的关系,为研究粗壮女贞的主要抑菌成分提供依据。方法不同极性溶剂对粗壮女贞水提物进行萃取,分光光度法分别测定不同萃取物的总黄酮和粗多糖的含量,通过微量肉汤稀释法测定提取物对不同细菌的最小抑菌浓度(MIC)。结果粗壮女贞水提物正丁醇萃取层对副溶血性弧菌、产气荚膜梭菌及拟杆菌的抑菌效果较好,最小抑菌浓度分别为:6.15g/L、13.2g/L、13.2g/L,剩余水层对副溶血性弧菌、嗜水气单胞菌及产气荚膜梭菌抑菌效果较好,最小抑菌浓度依次为:4.49g/L、10.7g/L和15.6g/L,三氯甲烷萃取层和乙酸乙酯萃取层对常见革兰阳性菌和革兰阴性菌几乎不抑制;正丁醇萃取层中的总黄酮含量最多,约占总水提物的85.6%,剩余水层中粗多糖的含量最多,约占总水提物的47.1%。结论粗壮女贞水提物对实验选择的几种致病菌有较好的抑制作用,对两种益生菌没有抑菌作用,抑菌活性物质可能主要为极性较大的物质,正丁醇萃取层和剩余水层的黄酮含量和粗多糖含量较高可能与其抑菌活性有关。Objective To study the diversity between general flavonoids and crude polysaccharides in different parts of extracts of Ligustrum robustum,and analyze its relationship with the difference of antibacterial activity,in order to provide the basis for the more study research on the antibacterial activity of Ligustrum robustum. Methods We used 80℃ water bath to get aqueous extracts of Ligustrum robustum,and then extracted the crude solution with three kinds of solvent,to divide crude extracts into four parts. In the following study,spectrophotometric methods were used to determine the quantity of flavonoids and soluble polysaccharides. And we determined the bacterial susceptibility with broth microdilution method. Results The n- butyl alcohol extracts showed it had high antimicrobial activity on Vibrio parahaemolyticus,Clostridium perfringens,and Bacteroides fragilis,and the Minimum Inhibition Concentration( MIC) were 6. 15 g / L,13. 2g / L,13. 2g / L,respectively. And the water remainders also inhibited the growth of Vibrio parahaemolyticus,Aeromonas hydrophilia and Clostridium perfringens,and the MIC were4. 49 g / L,10. 7g / L,15. 6g / L,respectively. Conclusion The results of bacterial susceptibility determination show that water extracts can selectively inhibit the growth of some pathogens and some intestinal bacteria. And the bioactive chemicals exist in the n- butyl alcohol extract and the remainders. Moreover,Most of the flavonoids and polysaccharides in the n- butyl alcohol extracts and the remainders might be the reason for the high antimicrobial performance,which still may need further study.
