摘 要:目的分析丹参(SMB)注射液对宫颈癌细胞的促增殖效应及其相关调控分子表达的影响。方法将人宫颈癌Hela细胞分别与终浓度为1.56 g/L、3.13 g/L、6.25 g/L、12.50 g/L、25.00 g/L、50.00 g/L、100.00 g/L、200.00 g/L的SMB共同孵育,噻唑蓝(MTT)法检测细胞生长增殖的A492值,流式细胞计数(FCM)分析细胞周期各时相细胞百分率及凋亡指数(AI)、增殖指数(PI);免疫细胞化学染色显示PCNA、Bcl-2、Bax分子表达水平,计算表达阳性细胞百分率,并以图像分析系统定量分析阳性表达强度。结果 12.50~200.00 g/L的SMB作用后,Hela细胞的生长增殖呈递进性显著增高(均P〈0.05),且与SMB浓度间呈显著正相关(P〈0.01);Hela细胞G0/G1期比例显著降低,S、G2/M期比例及PI值显著升高(均P〈0.05),AI值变化无显著差异(P〉0.05)。同时,Hela细胞中PCNA、Bcl-2的阳性表达率及表达强度明显增高,Bax的阳性表达率及表达强度明显降低,其变化均与SMB作用浓度呈显著相关关系。结论 SMB注射液可影响Hela细胞增殖凋亡调控分子的表达,发挥促增殖效应,在疾病治疗应用中须慎重。Objective To analyze the promoting effect of Salvia Miltiorrhiza Bunge( SMB) on the proliferation of cervical cancer cells and expressions of their regulators. Methods Hela cells( human cervical cancer cells) were co-incubated with SMB injection at different final concentrations( 1. 56 g / L,3. 13 g / L,6. 25 g / L,12. 50 g / L,25. 00 g / L,50. 00 g / L,100. 00 g / L,200. 00 g /L respectively). The proliferation of Hela cells was observed and values of A492 were measured by MTT method. Their cell cycles,apoptosis index( AI) and proliferation index( PI) were calculated through flow cytometry( FCM). Expressions of PCNA,Bcl-2 and Bax in Hela cells were analyzed through immunocytochemical staining,percentages of positive cells were calculated,and the intensity of positive staining was measured by image analysis system synchronously. Results SMB accelerated the growth and proliferation of Hela cells along with the increased concentrations ranged from 12. 50 g / L to 200. 00 g / L( all P 〈 0. 05),and their positive correlation was shown significantly( P 〈 0. 01). And the proportion of cells in stage of G0/ G1 decreased,that in S and G2/ M and values of PI elevated greatly( all P 〈 0. 05),but no significant changes among values of AI were shown( P 〉 0. 05). Meanwhile,after administered with SMB,the percentage and the intensity of PCNA and Bcl-2 positive expressions in Hela cells were higher,and those of Bax were lower greatly. The correlations were also significantly shown among changes of regulators’ expressions and concentrations of SMB. Conclusion SMB injection can regulate the expressions of proliferating regulators and promote the proliferation of Hela cells. SMB should be used cautiously and thoughtfully for the patients.
