磷酸神经鞘氨醇在人结肠癌中的表达及其判断结肠癌预后的研究

摘　　要：目的利用人结肠癌标本检测磷酸神经鞘氨醇(sphingosine 1-phosphate,s1p)及其相关激酶sphk1、sphk2的表达,研究其作为血清标记物判断结肠癌预后肿瘤标志物的价值。方法利用real-time PCR检测s1p及其相关激酶sphk1、sphk2的表达,免疫组织化学染色检测蛋白的表达及定位,并利用线性相关性分析及Kaplan-Meier曲线分析结肠癌患者血清s1p与预后的关系。结果 s1p及其受体相关激酶在人结肠癌中表达增高;sphk1在结肠癌组织中高表达;sphk2在正常组织中高表达,在结肠癌组织中表达下降;s1p的表达与sphk1呈正相关,而与sphk2的表达呈负相关;血清s1p的表达与患者的预后呈负相关。结论 s1p在结肠癌中高表达,是判断结肠癌患者术后差的生物标志物。Objective To investigate the expression of sphingosine 1- phosphate（ s1p）,and the expressions of the related kinases sphk1 and sphk2 in human colon cancer,and to study the value of serum marker in judging the prognosis of colon cancer.Methods The expressions of s1 p and its related kinases sphk1 and sphk2 in human colon cancer were determined by real-time PCR,and the distribution and protein localization were determined by immunohistochemistry,and the relation between serum s1 p and prognosis of post- surgery gastric patients was analyzed by Kaplan- Meier curve. Results Expressions of s1 p and its related kinases in human colon cancer increased; sphk2 highly expressed in normal issues and decreased in expression in colon cancer; The expression of s1 P was positively correlated with sphk1,but negatively correlated with the expression of sphk2.Serum s1 p was negatively related to prognosis of post- surgery cancer patients. Conclusion s1 p was overexpressed within human colon cancer,and can serve as an indicator of poor prognosis of post- surgery gastric cancer patients.
